摘  要:由于近年来臌囊苔草(Carex schmidtii Meinsh)湿地受到严重破坏,采用组织培养快繁可以有效解决这一问题,因此以臌囊苔草根茎为外植体,进行不同浓度次氯酸钠和升汞的单因素试验;在此基础上,开展外植体、次氯酸钠消毒时间和升汞消毒时间的正交试验。结果表明:适宜根茎外植体的消毒浓度分别为10%次氯酸钠和0.1%升汞;根茎的污染率和死亡率最低,存活率最高,为适宜的外植体。最佳消毒方式为75%的酒精浸泡30s,10%次氯酸钠消毒10min,0.1%升汞消毒5min,该方法污染率最低、存活率最高,分别为10.00%、71.11%。Carex schmidtii wetland in Sanjiang Plain were severely disturbed and destroyed because of exploitation of wetland resources.To solve this problem,we studied the tissue culture of Carex schmidtii plants.The disinfection effect of NaClO and HgCl2 for Carex schmidtii rhizome,and on this basis,we design orthogonal experiment with explants,disinfection time of NaClO and HgCl2 respectively.The results showed that in the single factor experiment with rhizome as explants,the concentrations of NaClO and HgCl2 with good disinfection effect were 10%and 0.10%respectively.In the orthogonal test,rhizome,compared with stem segments and leaves,had low pollution rate and high survival rate.The best disinfection way was that the explants were immersed in alcohol(75%)for 30s,NaClO(10%)for 10 min and then HgCl2(0.10%)for 5 min with the lowest pollution rate(10%)and highest survival rate(71.11%).
