摘 要:为探明解淀粉芽孢杆菌(Bacillus amyloliquefaciens)gfj-4次级代谢产物及其和化学杀菌剂的复配剂对葡萄白腐病菌(Coniella diplodiella)的抑制活性。采用菌丝生长速率法测定发酵上清液、脂肽粗提物以及发酵上清液与化学杀菌剂的混配剂对病菌的抑制特性。结果表明,种子液培养时间为4h与发酵培养时间为84h对病菌的抑制率最高。拮抗菌发酵上清液和脂肽粗提物对病菌的EC_(50)分别为11.10μL/mL和3.02μL/mL。葡萄白腐病菌对苯醚甲环唑、腈菌唑和戊唑醇的敏感性较高。其中,苯醚甲环唑对病菌的抑制活性最高,EC_(50)为0.02μg/mL。保护性杀菌剂福美双和百菌清对病菌抑制活性较高,EC_(50)分别为2.81μg/mL和3.51μg/mL。解淀粉芽孢杆菌gfj-4发酵上清液和苯醚甲环唑混配对葡萄白腐病菌的抑制活性表现为增效作用,其中与苯醚甲环唑以3∶7的体积比混配毒性比最高,达1.60。解淀粉芽孢杆菌gfj-4发酵上清液、脂肽粗提物以及发酵上清液与苯醚甲环唑的混配剂对葡萄白腐病菌有较强的抑制活性,具有潜在的利用价值。This study was conducted to investigate the inhibitory activity of the secondary metabolites of Bacillus amyloliquefaciens gfj-4 and its mixture with chemical fungicides against C.diplodiella.The antifungal activity of the fermentation supernatant,lipopeptide crude extract and the mixture of fermentation supernatant and fungicides were determined by mycelial growth rate method.The result showed that the most suitable incubation time of the seed liquid was 4 h and the appropriate fermentation time was 84 h.When the concentration of fermentation supernatant of B.amyloliquefaciens increased from 6.7μL/mL to 33.3μL/mL,the inhibition rate was between 16.7%and 90.0%,the EC 50 of the fermentation supernatant was 11.10μL/mL for C.diplodiella.When the lipopeptide crude extract increased from 1.0μL/mL to 16.0μL/mL,the inhibition rate was between 6.3%and 98.0%,the EC 50 of the lipopeptide crude extract was 3.02μL/mL for C.diplodiella.The pathogen was highly sensitive to ergosterol biosynthesis inhibitors difenoconazole,myclobutanil and tebuconazole.Among them,difenoconazole had the highest inhibitory activity against C.diplodiella with EC 50 of 0.02μg/mL.The EC 50 of the protective fungicides thiram and chlorothalonil were 2.81μg/mL and 3.51μg/mL,respectively.The B.amyloliquefaciens fermentation supernatant and difenoconazole mixed with the main performance of the additive effect reached the highest toxicity ratio of 1.60 when the ratio was 3∶7.B.amyloliquefaciens fermentation supernatant,lipopeptide crude extracts and the mixture of fermentation supernatant and difenoconazole had a strong inhibitory activity against grape white rot fungi and it had the potential value.
