摘  要:建立大鼠毛囊干细胞的体外无血清培养体系。使用化学成分确定的Knock Out血清替代物(KSR)代替现有毛囊干细胞生长培养基中的胎牛血清,比较两种培养基培养的毛囊干细胞的形态特点和生长情况;通过毛囊重建实验证明该无血清培养基能够在体外维持毛囊干细胞的"干性";检测无血清培养基培养的毛囊干细胞中α6-整合素、角蛋白K14和P63的表达情况。毛囊干细胞在该无血清培养基中可正常生长并表达上述蛋白,将其移植到裸鼠背部,能够重建含毛囊皮肤。建立了大鼠毛囊干细胞的体外无血清培养体系并重建有毛皮肤,为毛囊干细胞的临床研究和应用提供了理论基础和技术支持。To establish a serum-free rat hair follicle stem cells(HFSCs)culture system.Fetal bovine serum(FBS)in the currently used HFSCs growth medium was replaced by chemically defined KnockOut serum replacement(KSR).Cell morphology and growth curve were compared between HFSCs cultured in the serum-free culture medium and FBS-containing culture medium.Hair follicle reconstitution experiment was conducted to validate whether stemness of HFSCs cultured in the serum-free culture medium was maintained.In addition,the expression ofα6-intergin,K14 and P63 in HFSCs cultured in the serum-free culture medium was detected by immunofluorescence staining.HFSCs cultured in the serum-free culture medium showed normal cell morphology and growth rate.Hair follicle-containing skin was reconstituted by HFSCs cultured under serum-free culture medium.Proteins includingα6-intergin,K14 and P63 highly expressed in HFSCs were still expressed in HFSCs cultured in serum-free culture medium.Serum-free rat HFSC culture medium was established.This provides the theoretical basis and technical support for the clinical research and application of HFSCs.
