摘 要:目的探讨4-羟苯基维胺(4-HPR)对瘢痕疙瘩成纤维细胞Ⅰ型前胶原(ProcollagenⅠ)和基质金属蛋白酶-1(MMP-1)mRNA表达的影响。方法运用实时荧光定量PCR方法测定瘢痕疙瘩成纤维细胞中ProcollagenⅠ和MMP-1 mRNA表达情况。观察4-HPR对ProcollagenⅠ和MMP-1 m RNA表达的影响。结果瘢痕疙瘩成纤维细胞中ProcollagenⅠmRNA表达明显高于正常皮肤成纤维细胞,而MMP-1 mRNA低表达,差异具有统计学意义。4-HPR不同浓度组瘢痕疙瘩成纤维细胞ProcollagenⅠmRNA表达水平明显减少,MMP-1 mRNA表达水平明显增加,差异具有统计学意义。结论 ProcollagenⅠ和MMP-1基因表达在瘢痕生成过程中起重要作用。4-HPR可抑制ProcollagenⅠ和促进MMP-1 mRNA表达水平。Objective To investigate the effect of 4-HPR onProcollagenⅠand MMP-1 mRNA expression in keloid fibroblasts.Methods The expression ofProcollagenⅠand MMP-1 mRNA was detected by real-time PCR in keloid fibroblasts.The expression of ProcollagenⅠand MMP-1 mRNA was treated by 4-HPR in keloid fibroblasts.Results The expression of ProcollagenⅠmRNA in keloid fibroblasts was significantly higher than that in the normal fibroblasts,while MMP-1 mRNA was significantly lower.Expression of ProcollagenⅠmRNA decreased and MMP-1 mRNA increased in 4-HPR group.Conclusion ProcollagenⅠand MMP-1 may play some roles in the regulation of keloid formation.4-HPR inhibits ProcollagenⅠexpression and promotes MMP-1 mRNA expression in keloid fibroblasts.
