摘 要:目的探讨组蛋白去乙酰化酶9(HDAC9)基因rs3757720单核苷酸多态性(SNP)与冠心病阳性相关性。方法选取自2014年6月至2017年4月沈阳军区总医院收治的1 101例冠状动脉疾病(CAD)患者作为冠心病组,同期体检的891例无心血管疾病的健康人作为健康组。采用连接酶检测反应测定HDAC9基因SNP rs3757720的基因分型,比较两组等位基因的频率以及HDAC9 SNP与CAD发生风险之间的等位基因和基因型关联分析。结果冠心病组中rs3757720 T等位基因频率为10. 9%,显著高于健康组的4. 5%(OR=1. 234,95%可信区间1. 147~1. 327,P=0. 025)。调整了潜在的混杂因素,如年龄、性别、高血压、糖尿病、吸烟状况及血脂水平后,CAD与A等位基因携带者相比较(OR=2. 344,95%可信区间2. 051~2. 523,P=0. 019),携带TT纯合子和AT杂合子的受试者易感性显著增加。结论 HDAC9基因的SNP rs3757720与CAD的发生有独立相关性,HDAC表达增加可能对TT+AT基因型的CAD的发生具有促进作用。Objective To investigate the positive correlation between histone deacetylase 9(HDAC9)single nucleotide polymorphism(SNP)rs3757720 and coronary heart disease.Methods A retrospective study was performed on 1 101 cases of patients with coronary artery disease(CAD)who were admitted from June 2014 to April 2017.All the 1 101 cases of CAD patients were divided into the CAD group,meanwhile,the other 891 cases of healthy volunteers without CAD were in the healthy group.The genotypes of HDAC9 single nucleotide polymorphism(SNP)rs3757720 were determined by ligase assay,and the alleles and genotype correlations between HDAC9 SNP and CAD risk were compared and analyzed.Results The rs3757720 T allele frequency was 10.9%in CAD group,which was significantly higher than 4.5%in healthy group(OR=1.234,95%CI 1.147 to 1.327,P=0.025).After adjusting for potential confounders,such as age,gender,hypertension,diabetes,smoking status and lipid levels,CAD was significantly increased compared to A allele carriers(OR=2.344,95%CI 2.051 to 2.523,P=0.019),subjected with TT homozygous and AT heterozygous significantly increased susceptibility.Conclusion SNP rs3757720 of HDAC9 gene is independently associated with the development of CAD,and TT+AT genotype which is associated with increased expression of HDAC may have a promoting effect on CAD.
