摘 要:本试验旨在研究中链脂肪酸对脂多糖(LPS)诱导的断奶仔猪肠黏膜免疫屏障损伤的保护作用。将24头(21±1)d、体重为(9.0±0.7)kg的杜×长×大三元杂交断奶仔猪随机分为4个处理组,每个处理组6个重复,每个重复1头猪。试验采用2×2因子设计,2个主因子分别为日粮处理(中链脂肪酸或玉米油)和免疫应激(LPS或NaCl)。预试7 d,试验期为21 d。试验第21天,每个日粮组3个重复注射100μg/kg体重(BW)的LPS,另外3个重复猪注射等量的生理盐水,4 h后屠宰取肠道样品待测。结果表明:中链脂肪酸显著提高了空肠绒毛高度(P<0.05),降低了空肠、回肠固有层细胞密度和回肠PP结细胞密度(P<0.05);中链脂肪酸还缓解了LPS刺激导致的空肠杯状细胞数的升高和上皮间淋巴细胞数的减少。综上,日粮中添加中链脂肪酸有保护仔猪肠黏膜免疫屏障的功能。The experiment was conducted to investigate the effects of medium-chain fatty acids(MCFA)supplementation on intestinal mucosal immune barrier function of weanling piglets after lipopolysaccharide(LPS)challenge.A total of 24 weaned piglets of(21±1)d with average body weight(9.0±0.7)kg,were randomly divided into four treatments.All piglets fed control diet for 7 d while they were acclimated to the experimental conditions.The experiment consisted of a 2×2 factorial design,and the main factors included diet(MCFA or corn oil)and immunological challenge(LPS or sterile saline).On d 21 of the trial,pigs were injected with 100μg/kg BW of LPS or saline.At 4 h after sterile saline or LPS administration,pigs were humanely killed and intestinal samples were collected.MCFA supplementation maintained the integrity of intestinal morphology by increase the height of villi in jejunum(P<0.05).Pigs fed MCFA had lower density of lamina propria cell and peyers pathch(PP cells)in ileum than piglets fed corn oil(P<0.05).MCFA supplementation alleviated the increase of the number of goblet cells and the decrease of the number of intraepithelial lymphocytes induced by LPS challenge.These results indicated that dietary MCFA can improve intestinal mucosal morphology and protect intestinal barrier function.
