摘 要:以盐芥为试验材料,研究不同浓度镉胁迫下盐芥根木栓质组分及含量、木栓质代谢相关基因表达变化,旨在分析盐芥根木栓质对不同浓度镉胁迫的响应。结果表明,2种浓度(50μmol/L、100μmol/L)镉胁迫下,盐芥根木栓质优势组分未发生变化,检测出的4种类型的木栓质,其含量从高到低依次为ω-羟基脂肪酸、未被取代脂肪酸、α,ω-二羧酸、脂肪醇; 4种类型的木栓质包括21种单体,50μmol/L镉胁迫下,木栓质总量升高,而100μmol/L镉胁迫下,木栓质总量未发生明显变化;对木栓质代谢相关的16个基因进行表达模式分析,50μmol/L镉胁迫下,CYP86B1、CYP86A7、FAR4、FAR5、KCR2、ABCG2、GPAT6、FACT、MYB107、CYP86A4、KCR20、ABCG6、ABCG20、GPAT5、ASFT共15个基因显著或极显著上调表达,100μmol/L镉胁迫下,CYP77A6、CYP86A7、FACT、CYP86A4、ABCG2、GPAT5、ASFT、MYB107共8个基因显著或极显著上调表达。Root suberin component and its content,suberin metabolism related genes expression were assayed in Thellungiella salsuginea under cadmium stress,aimed to explore the effect of cadmium stress on suberin metabolism.The results showed that the dominant components of root suberin were not changed under cadmium stress(50μmol/L,100μmol/L),which wereω-hydroxyacids,unsubstituted fatty acids,α,ω-dioicids and fatty alcohols according to the content from high to low.21 suberin monomers were detected in the roots of Thellungiella salsuginea.The total amount of suberin monomers increased under 50μmol/L cadmium,while did not significantly change under 100μmol/L cadmium.Under 50μmol/L cadmium stress,there were 15 genes(CYP86B1,CYP86A7,FAR4,FAR5,KCR2,ABCG2,GPAT6,FACT,MYB107,CYP86A4,KCR20,ABCG6,ABCG20,GPAT5 and ASFT)which were significantly or extremely significantly up-regulated.Under 100μmol/L cadmium stress,there were 8 genes(CYP77A6,CYP86A7,FACT,CYP86A4,ABCG2,GPAT5,ASFT and MYB107)which were significantly or extremely significantly up-regulated.
