摘  要:目的研究丙泊酚对N-甲基-D-天冬氨酸(NMDA)毒性损伤的保护作用及其机制。方法采用昆明小鼠制备离体海马脑片,随机分为对照组、损伤组和丙泊酚组(1、5、10μmol/L),每组10张脑片。2,3,5-氯化三苯四氮唑(TTC)染色法测定NMDA毒性损伤后脑片组织损伤率。细胞外记录技术观察顺向群峰电位(OPS)的变化。免疫组织化学方法观察海马CA1区脑源性神经营养因子(BDNF)蛋白表达的变化。结果与损伤组比较,丙泊酚(5、10μmol/L)可明显减轻NMDA造成的脑片组织损伤(P<0.01),提高海马脑片OPS恢复程度(P<0.01),明显增强BDNF表达(P<0.05或P<0.01)。结论丙泊酚可通过增强BDNF的表达,减轻细胞损伤,促进神经元突触传递功能的恢复。Objective To investigate the protective effects of propofol on toxic injury induced by N-methyl-D-aspartate(NMDA)and the relative mechanisms.Methods Hippocampal slices were prepared from Kunming mice and randomly divided into the control group,the injury group and the propofol group(1,5,10μmol/L),10 slices per group.The injury of hippocampal slices was measured by 2,3,5-triphenyltetrazolium chloride(TTC)staining.The changes of orthodromic population spike(OPS)after toxic injury induced by NMDA were observed by extracellular recording technique.The expression of brain-derived neurotrophic factor(BDNF)in hippocampal CA1 regions was measured by immunohistochemical assay.Results Compared with the injury group,propofol(5,10μmol/L)significantly attenuated hippocampal tissue injury induced by NMDA(P<0.01),improved the recovery amplitude of OPS(P<0.01),and increased the expression of BDNF in hippocampal CA1 regions(P<0.05 or P<0.01).Conclusion Propofol could antagonize NMDA toxicity and improve the recovery of synaptic transmission neurons,which might be related to the upregulation of BDNF expression.
