电针联合健中愈疡片对实验性胃溃疡模型大鼠β-内啡肽及炎症因子的影响

摘　　要：目的:观察电针联合健中愈疡片对实验性胃溃疡模型大鼠β-内啡肽及炎症因子的影响,探讨针药结合治疗胃溃疡的机制。方法:采用乙酸诱导制备大鼠胃溃疡模型。造模后将50只SD大鼠随机分为假手术组、模型组、电针组、药物组和联合组,每组10只,其中假手术组大鼠未进行乙酸滤纸刺激,其他操作相同。电针组大鼠采用持续脉冲电流,频率2 Hz、强度2 m A针刺足三里和梁门穴;药物组灌胃给予健中愈疡片0. 70 g·kg^(-1);联合组同时采用电针与健中愈疡片治疗;假手术组和模型组给予等量生理盐水。连续治疗10 d后,采用酶联免疫吸附法检测血浆和下丘脑中β-内啡肽,血清中炎症因子白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)的含量。结果:与假手术组比较,模型组大鼠胃溃疡损伤指数、下丘脑β-内啡肽含量和血清中IL-6、IL-8、TNF-α含量明显上升(P <0. 05),血清β-内啡肽含量降低;与模型组比较,电针组、药物组和联合组胃溃疡损伤指数、下丘脑β-内啡肽含量和血清中IL-6、IL-8、TN F-α含量明显降低,血清β-内啡肽含量上升(P <0. 05);联合组胃溃疡损伤指数、下丘脑β-内啡肽和血清中炎症因子表达显著低于电针组或药物组(P <0. 05)。结论:电针与健中愈疡片结合能够通过降低血浆β-内啡肽水平调节免疫功能,抑制炎症反应治疗胃溃疡,疗效优于单独使用电针或健中愈疡片。Objective:To observe the effect of electroacupuncture combine Jianzhongyuyang Pill onβ-endorphin and inflammatory factor in experimental gastric ulcer model rats.Methods:Acetic acid was used to induce gastric ulcer model rats.After modeling,fifty SD rats were randomly divided into sham-operated group,model group,electroacupuncture group,medicine group,combined group(n=10).The rats in the sham-operation group did not receive acetic acid filter paper stimulation,and the other operations were the same.Continuous pulse current was applied to Zusanli and Liangmen of rats in electroacupuncture group with frequency of 2 Hz and intensity of 2 mA.Jiangzhongyuyang Pill was given by gavaging to rats in medicine group(0.70 g·kg-1).Electroacupuncture and Jiangzhongyuyang Pill were adopted at the same time to rats in combined group.Saline was given by gavaging to rats in sham-operated group and model group.Content ofβ-endorphin in plasma and hypothalamus,content of interleukin-6(IL-6),interleukin-8(IL-8),tumor necrosis factor-α(TNF-α)in serum were detected by enzyme linked immunosorbent assay.Results:Compared to the sham-operated group,content of gastric ulcer injury index,β-endorphin in hypothalamus and IL-6,IL-8,TNF-αin serum were significant increase and theβ-endorphin in plasma was significantly reduced(P<0.05).Compared to the model group,content of gastric ulcer injury index,β-endorphin in hypothalamus and IL-6,IL-8,TNF-αin serum were significant decreased and theβ-endorphin in plasma was significantly increased(P<0.05).Content of gastric ulcer injury index,β-endorphin in hypothalamus and IL-6,IL-8,TNF-αin serum of combined group rats were lower than those in electroacupuncture group or medicine group.Conclusion:Electroacupuncture plus Jianzhongyuyang Pill is able to cure gastric ulcer by decreasing level ofβ-endorphin in plasma,results in inflammatory response inhibiting,and its effects are apparently superior to those of simple electroacupuncture or Jianzhongyuyang Pill.
