摘 要:目的:探究不同运动方式对大鼠比目鱼肌内质网应激的影响。方法:32只雄性SD大鼠随机分成安静对照组(C)、一次性力竭运动组(L)、6周运动组(Y)、6周游泳运动后一次性力竭运动组(LY),每组8只。6周游泳训练大鼠无负重自由游泳,每周训练6次(周日休息一天),每次训练60min,末次训练结束后禁水禁食,休息24h,次日断头取比目鱼肌。一次性游泳力竭标准采用3%体重负重游泳至大鼠下层水底10s不露出水面,大鼠游至力竭后捞出,自然恢复40min,再放入池内,游至力竭,运动后即刻断头取比目鱼肌。BCA法测定比目鱼肌全蛋白含量;Western-blot法测定比目鱼肌GRP78和CHOP的蛋白表达。结果:一次性力竭组GRP78表达水平显著高于有氧运动组(P <0.05)和安静对照组(P <0.05),其余各组之间无显著差异,各组间大鼠比目鱼肌CHOP表达水平无显著性差异。结论:一次性游泳力竭运动诱发比目鱼肌内质网应激;6周游泳训练未引起内质网应激的发生;长期游泳训练后,比目鱼肌内质网对力竭运动适应性增强;内质网应激相关细胞凋亡指标CHOP的表达变化可能具有滞后性。Objective:To investigate the effect of different modes of exerise on endoplasmic reticulum stress in skeletal muscle of rats.Methods:Thirty-two male SD rats were randomly divided into four groups of 8 in each group:A quiet control group(C),one-time exhaustive exercise group(L),6-week swimming group(Y),and onetime exhaustive exercise group after 6 weeks swimming(LY).The 6-week swim training rats had no weightbearing free swimming,training 6 times a week(one day off on Sunday),and each training for 60 minutes.Onetime swimming exhaustion criteria:3%of body weight weight-bearing swimming,swimming to rats sinking into the bottom of the pool,and within 10s does not reveal the surface of the water.The rats swim until exhaustion and naturally recover 40min,then put into the pool again.The rats were allowed to swim until exhaustion.The rats were immediately decapitated after exercise.Determination of total protein content of soleus muscle by BCA;the protein expression of GRP78 and CHOP in soleus muscle was tested by Western-blot.Results:The expression level of GRP78 in the group L was significantly higher than that in the group L(P<0.05)and group C(P<0.05).Conclusion:One-time swimming exhaustion induced endoplasmic reticulum stress in soleus muscle;6 weeks of swimming training did not cause endoplasmic reticulum stress;after long training swimming training,the adaptability of the soleus endoplasmic reticulum to exhaustion movement was enhanced,and the expression of CHOP expression in endoplasmic reticulum stress related apoptosis index may be delayed.
