摘 要:为了深入研究百合响应高温胁迫的分子机制,本研究以麝香百合杂种系品种白天堂为试验材料,从中分离得到1个MBF1c基因,其开放阅读框为438 bp,编码145个氨基酸,蛋白质分子量为15 900,理论等电点为10. 22。多序列同源比对和进化树分析表明百合MBF1c具有典型的MBF1和HTH结构域,归属c亚型MBF1,因此将克隆到的基因命名为LlMBF1c。亚细胞定位结果表明,LlMBF1c可能介导了热激信号由细胞质向细胞核的转导。实时荧光定量PCR分析结果表明,LlMBF1c在叶中的表达量最高,根中次之,鳞茎中最低; LlMBF1c的表达受热激诱导。表明,LlMBF1c基因可能与百合的热激反应密切相关。To explore the molecular mechanism of lily in response to heat stress,the MBF1c homologous gene was isolated from lily cultivar White Heaven.The open reading frame of LlMBF1c gene was 438 bp,a protein of 145 amino acid residues was encoded,and the molecular weight of LlMBF1c protein was 15 900 with a theoretical isoelectric point of 10.22.Sequence alignment and phylogenetic relationship analysis results showed that LlMBF1c belonged to c subtype MBF1,containing a typical MBF1 domain and a HTH domain.Subcellular localization assay results indicated that LlMBF1c could mediate the transduction of heat stress signal from cytoplasm to nucleus.The qRT-PCR results showed that the relatively highest expression level of LlMBF1c was detected in leaf and LlMBF1c transcripts were induced by heat shock.In conclusion,LlMBF1c might be closely related to heat stress response in lily.
