摘 要:目的探讨内质网应激对胃癌SGC-7901细胞凋亡的影响。方法将胃癌SGC-7901细胞根据不同的处理方式分为3组,即对照组(Control)、内质网应激组(ERS)和内质网抑制剂组(4-PBA),其中内质网应激组用浓度为3.5μmol/L的衣霉素(tunicamycin,TM)处理,诱导内质网应激;抑制剂组用100 nmol/L的4-苯基丁酸(4-phenyl butyric acid,4-PBA)处理,抑制内质网应激。各组细胞处理24 h后收集细胞,采用MTT法检测细胞的活性,流式细胞仪检测细胞的凋亡率,Western blot法检测免疫球蛋白结合蛋白(binding immunoglobulin protein,BIP)、CCAAT增强子结合蛋白同源蛋白(C/EBP homologus protein,CHOP)、凋亡特异性蛋白(Caspase-3)的表达水平。结果与对照组比较,内质网应激组的细胞活性明显下降(P <0.01),细胞凋亡率增加(P <0.01),Bip、CHOP及Caspase-3蛋白表上调(P <0.05);而抑制剂组差异无统计学意义(P> 0.05)。结论衣霉素可诱导胃癌SGC-7901细胞内质网应激并上调Caspase-3蛋白表达水平,引起胃癌细胞凋亡。Objective To explore the effect of apoptosis for endoplasmic reticulum stress in gastric cancer SGC-7901 cells.Methods The SGC-7901 cells were divided into 3 groups according to the different treatments:control group(Control),endoplasmic reticulum stress group(ERS),and inhibitor group(4-PBA).endoplasmic reticulum stress group was induced endoplasmic reticulum stress by treated with tunicamycin(TM)with a concen-tration of 3.5μmol/L,and inhibitor group was treated with 4-phenyl butyric acid(4-PBA)with a concentration of 100 nmol/L.When these cells were cultured for 24 h,the cell activity was assessed by the MTT staining.The cell apoptosis was determined by the fluorescence microscopy analysis after Hoechst/PI staining and flow cytometric assay after Annexin V-FITC/PI staining.The expressions of the Bip(binding immunoglobulin protein),CHOP(C/EBP homologus protein),and Caspase-3 protein were detected by the Western blot.Results Compared with the control group,the cell activities were significantly decreased(P<0.01),the cell apoptosis rates and the Bip,CHOP and Caspase-3 protein expressions were significantly increased(P<0.05)of the endoplasmic reticulum stress group,but in inhibitor group,the difference was not statistically significant(P>0.05).Conclusion Tu-nicamycin can induce the endoplasmic reticulum stress and increase the expression level of Caspase-3 protein in gastric cancer SGC-7901 cells,and causes gastric cancer cells apoptosis.
