摘  要:目的基于NF-κB/Bcl-2信号通路,研究富氢水对大鼠离体心肌缺血再灌注损伤细胞凋亡的影响。方法将48只大鼠随机分为对照组与富氢水组,每组24只,两组再分别随机分成缺血前期、缺血期、缺血再灌注期,每期8只大鼠。应用Langendroff灌流装置,对照组采用K-R液灌注,富氢水组采用K-R液+富氢水灌注验完毕后取材,TUNEL法检测各组心肌细胞凋亡率;采用免疫组织化学法测定各组心肌细胞Bcl-2、Bax、caspase3蛋白表达水平;Western Blot检测各组心肌细胞NF-κB蛋白表达。结果对照组与富氢水组缺血前期均无凋亡,缺血期凋亡明显;对照组缺血再灌注期与缺血期比较,心肌细胞凋亡明显增加(P <0.01);富氢水组缺血再灌注期与缺血期比较,差异无统计学意义;富氢水组缺血再灌注期与对照组缺血再灌注期比较,心肌细胞凋亡明显减少(P <0.01);对照组缺血期与富氢水组缺血期比较,差异无统计学意义。对照组缺血再灌注期与缺血期相比较,Bcl-2蛋白表达水平下降(P <0.05),Bax、caspase3蛋白表达水平增高(P <0.05);富氢水组缺血再灌注期与对照组缺血再灌注期相比,Bcl-2蛋白表达水平增高(P <0.05),Bax、caspase3蛋白表达水平降低(P <0.05)。对照组缺血再灌注期与缺血前期、缺血期比较NF-κB蛋白含量增高,差异有统计学意义(P <0.05);富氢水组缺血再灌注期与缺血前期、缺血期比较NF-κB蛋白含量增高,差异有统计学意义(P <0.05);富氢水组缺血再灌注期与对照组缺血再灌注期相比NF-κB蛋白含量明显降低(P <0.01)。结论富氢水能够通过调节NF-κB/Bcl-2通路减轻大鼠心肌缺血再灌注损伤,抑制心肌细胞凋亡。Objective Based on the signal pathway of NF-κB/Bcl-2,the effects of hydrogen-rich water on apoptosis of rat myocardial ischemia-reperfusion injury were studied.Methods A total of 48 rats were randomly divided into control group and hydrogen rich water group with 24 rats in each group.Two groups were randomly divided into pre ischemic period,ischemic period,ischemia reperfusion period with 8 rats in each period.Langen-droff perfusion device was used.The control group was treated with K-R perfusion and hydrogen rich water group was treated with K-R+hydrogen rich water.The TUNEL method was used to detect the apoptosis rate of cardiomyo-cytes in each group.The expression of Bcl-2,Bax and caspase3 protein in cardiomyocytes of each group was detected by immunohistochemical method.The expression of NF-κB protein in myocardial cells of each group was detected by Western Blot.Results Both the control group and the hydrogen-rich water group had no apoptosis during the pre-ischemic period.There was significant apoptosis in the ischemic period.Compared with the ischemic phase,the apoptosis of cardiac myocytes was significantly increased in the control group in ischemia reperfusion period(P<0.01).In the hydrogen enriched water group,the apoptosis of cardiac myocytes in ischemia-reperfusion group were significantly lower than those in control group(P<0.01).The expression of Bcl-2 protein in the control group was lower than that in the ischemia-reperfusion group(P<0.05),and the expression of Bax and caspase3 protein was increased(P<0.05).In the hydrogen enriched water group,the expression level of Bcl-2 protein increased(P<0.05)and the expression level of Bax and Caspase3 decreased(P<0.05)compared with the ischemia reper-fusion period in the control group.Compared with the pre ischemic stage and ischemic phase,the contents of NF-KB increased in the control group in ischemia reperfusion period(P<0.05).Compared with the pre ischemic stage and the ischemic stage,the contents of NF-κB increased in the hydrogen enriched water gro
