摘 要:目的探索NF-κB通路在自身免疫性甲状腺炎发病过程中的作用。方法将提取纯化的小鼠甲状腺球蛋白(mTg)联合弗氏佐剂免疫CBA/j小鼠建造实验性自身免疫甲状腺炎模型(EAT);利用HE染色检测病理变化;ELISA实验检测血清中TNF-α含量;qPCR实验检测甲状腺组织中TNF-α、Bcl-2、ICAM-1 3个蛋白的mRNA变化;Western bloting实验检测甲状腺组织中NF-κB蛋白的变化;免疫组化检测甲状腺组织中IKK蛋白的变化。结果成功提取mTg后,进行免疫造模,HE染色发现模型组甲状腺明显发炎,造模成功;与对照组相比,血清中TNF-α含量以及甲状腺组织中TNF-α、Bcl-2、ICAM-1的mRNA、NF-κB以及IKK蛋白均显著增加。结论自身免疫性甲状腺炎发病过程中,与TNF-α形成正反馈环导致NF-κB通路持续激活,启动Bcl-2和ICAM-1基因表达,促使炎症细胞增殖、浸润至甲状腺组织,促进AITD的发生发展。Objective To explore the role of NF-κB pathway in the pathogenesis of autoimmune thyroid-itis.Methods The purified mouse thyroglobulin(mTg)was combined with Freund′s adjuvant to immunize CBA/j mice to construct an experimental autoimmune thyroiditis model(EAT);HE staining was used to detect pathologi-cal changes;ELISA was used to detect TNF-αin serum.The content of TNF-α,Bcl-2 and ICAM-1 mRNA in thy-roid tissue was detected by qPCR assay.The changes of NF-κB protein in thyroid tissue were detected by Western blotting.The expression of IKK protein in thyroid tissue was detected by immunohistochemistry.Results After successful extraction of mTg,HE staining showed that the model group was significantly inflamed,demonstrating the model was successfully established.Compared with the control group,the level of TNF-αin the serum was sig-nificantly increased.The levels of TNF-α,Bcl-2,the mRNA,NF-κB and IKK proteins of three ICAM-1 proteins were significantly increased.Conclusion During the pathogenesis of autoimmune thyroiditis,a positive feedback loop with TNF-αleads to the continuous activation of the NF-κB pathway,which initiates the expression of Bcl-2 and ICAM-1 genes,promotes the proliferation and infiltration of inflammatory cells into thyroid tissue,and pro-motes the occurrence of AITD.
