摘  要:目的探讨microRNA-126(miR-126)在子痫前期孕产妇胎盘中的表达及其对血管生成的影响。方法选择子痫前期孕产妇50例(其中轻度子痫前期25例,重度子痫前期25例);随机选取年龄、孕周匹配的正常妊娠孕产妇25例作为对照。采用q RT-PCR检测胎盘组织中miR-126的表达水平。利用CD34抗体标记血管内皮细胞,计算胎盘组织中微血管密度。采用CCK-8试剂盒和Transwell小室装置检测细胞增殖和迁移,使用Matrigel胶模型观察小管形成。结果 miR-126在子痫前期组胎盘中的表达水平低于正常妊娠组(P <0.05)。miR-126表达状态与胎盘微血管密度相关(P <0.05)。体外细胞实验显示,下调miR-126表达可抑制血管内皮细胞增殖、迁移和小管形成能力(P <0.05)。结论 miR-126在子痫前期胎盘组织中的表达降低,可能通过调控血管生成参与子痫前期的发病。Objective To investigate the expression of miR-126 in placental tissue from preeclamptic patients,and study its effect on angiogenesis.Method The expression of miR-126 was determined by qRT-PCR.Microvascular density(MVD)assessment was based on CD34 expressing vessels.CCK-8,Transwell migration and Matrigel tube formation assay were performed to detect the effects of miR-126 on cell proliferation,migration and tube formation,respectively.Results miR-126 was decreased significantly in the placenta of patients with pre-eclampsia compared to the healthy controls(P<0.05).The expression of miR-126 was postively correlated with MVD in placentas(P<0.05).Down-regulation of miR-126 inhibited proliferation,migration and tube formation of HMEC-1 cells(P<0.05,respectively).Conclusion miR-126 might play a distinct role in the pathogenesis of preeclampsia by modulating angiogenesis.
