摘 要:为了探讨泡桐丛枝病相关蛋白质的表达变化情况,以白花泡桐组培苗为试验材料,采用iTRAQ技术对白花泡桐健康苗、患丛枝病苗进行了蛋白质组学分析。结果表明,在泡桐丛枝病发生过程中有1 788个蛋白质表达发生了显著变化,其中873个蛋白质的表达水平上升,915个蛋白质的表达水平下降。GO功能分析发现,差异蛋白质主要分为生物学过程、细胞组分和分子功能3个本体共49个功能组,功能主要集中在刺激响应、催化活性和生物学调控等方面。COG数据库进行比对结果表明,这些蛋白质主要参与碳水化合物的运输和代谢、翻译、转录、复制、重组和修复以及次生代谢物合成运输等生命活动。以KEGG数据库作为参考,依据代谢途径可将蛋白质定位到122个代谢途径分支,涉及次生代谢的生物合成、植物激素信号转导和植物—病原互作等。To investigate the expression variability of associated proteins in Paulownia witches’broom,iTRAQ-based quantitative proteomic technique was used to analyze proteomics of healthy and coniferous seedlings of Paulownia fortunei.The results showed that a total of 1788 differentially expressed proteins were identified during the occurrence of Paulownia witches’broom,among with 873 proteins expression levels increased,while 915 proteins expression levels decreased.All assembled proteins can be broadly divided into cellular components,molecular function and biological processes categories of 49 branches by gene ontology,including stimuli-responsive,catalytic activity and biological regulation.Proteins were annotated based on COG category,which participated in life activities such as carbohydrate transport and metabolism,translation,transcription,replication,recombination and repair,secondary metabolites biosynthesis,transport and catabolism.KEGG pathway analysis indicated that the proteins can be broadly divided into 122 classes according to the functions,including biosynthesis of secondary metabolites,plant hormone signaling and plant-pathogen interaction and the like.
