摘  要:目的观察神经生长因子(NGF)及其低亲和力受体(p75NTR)对大鼠肝星状细胞(HSC)凋亡及增殖的影响以及大鼠HSC中相关凋亡蛋白变化,探索NGF影响HSC凋亡的可能作用机制。方法体外培养大鼠肝星状细胞(HSC-T6),将HSC-T6与不同浓度的NGF及NGF+P75NTR孵育,分为对照组、NGF组和NGF+P75NTR组,对照组采用0 ng/ml NGF+0 ng/ml p75NTR,NGF组采用100 ng/ml NGF,NGF+p75NTR组采用100 ng/ml NGF+100 ng/ml p75NTR,应用流式细胞术对HSC凋亡情况进行检测,通过XTT法了解NGF及p75NTR对HSC增殖的影响;采用免疫细胞化学法观察NGF及p75NTR对HSC作用后P53、Caspase-3表达的变化。结果 NGF组对HSC的增殖抑制与NGF+p75NTR组及对照组比较,差异均有统计学意义(P<0.05)。流式细胞术检测结果显示,对照组的凋亡率低于NGF组和NGF+P75NTR组,差异有统计学意义(P<0.05),NGF+p75NTR组较NGF组凋亡率明显升高(P<0.05)。NGF组和NGF+P75NTR组的P53、Caspase-3表达与对照组比较明显升高,差异有统计学意义(P<0.05),其中NGF+p75NTR组升高较NGF组明显(P<0.05)。结论 NGF可能通过p75NTR抑制HSC-T6增殖、促进其凋亡,其机制可能与调节P53、Caspase-3表达有关。Objective To observe the effect of nerve growth factor(NGF)and its low affinity receptor(p75 neurotrophin receptor,p75NTR)on the apoptosis and proliferation of rat hepatic stellate cells(HSC),and to explore the possible mechanism of apoptosis.Methods Rat hepatic stellate cells(HSC-T6)were incubated in vitro,HSC-T6 was incubated with NGF and NGF+p75NTR with different concentrations.The cells were divided into the control group,the NGF group and the NGF+p75NTR group.The control group was cultured with 0 ng/ml NGF+0 ng/ml p75NTR,the NGF group was cultured with 100 ng/ml NGF,and the NGF+p75NTR group was cultured with 100 ng/ml NGF+100 ng/ml p75NTR.The flow cytometry was used to detect the apoptosis condition.The effect of HSC on the proliferation of NGF and p75NTR was observed by XTT,the expression of P53 and Caspase-3 were detected by immunohistochemical staining.Results The inhibitory effect of the NGF group on the proliferation of HSC was statistically significant(P<0.05)with the NGF+p75NTR group and the control group(P<0.05).The flow cytometry results showed that the apoptosis rate of the control group was lower than that of the NGF group and the NGF+p75NTR group(P<0.05),the apoptosis rate of the NGF+p75NTR group was higher than the NGF group(P<0.05).The expression of P53 and Caspase-3 in the NGF group and the NGF+p75NTR group were significantly higher than that in the control group(P<0.05),the expressions in the NGF+p75NTR group were significantly higher than that in the NGF group(P<0.05).Conclusion NGF may inhibit HSC-T6 proliferation and promote its apoptosis through p75NTR,which may be related to the regulation of P53 and Caspase-3 expression.
