摘  要:目的对四川省鼠疫耶尔森菌(Yersinia pestis)做规律成簇的间隔短回文重复序列(Clustered regularly interspaced short palindromic repeats CRSIPR)基因分型和地区分布研究,为四川鼠疫防治提供科学依据。方法对132株鼠疫菌培养和提取核酸,应用3对CRISPR引物对被试菌株DNA进行PCR扩增、测序和分析,确定四川省鼠疫菌的CRSIPR基因型。结果 132株鼠疫菌共发现17种spacer,包括YPa 9种、YPb 5种、YPc 3种,新发现的1种space阵列。所有鼠疫菌株被分为2个CRISPR基因簇(Cc3′,Ca7),3个基因型(37′型、22型和sc01型)。其中石渠县青海田鼠型菌株基因型为37′型,石渠县人间鼠疫的菌株为22型,德格县旱獭型菌株基因型为22型,巴塘县、理塘县、雅江县和新龙县旱獭菌株基因型为sc01型。结论四川省鼠疫菌菌株具有明显的地区分布特征。We studied the CRISPR genotypes data and regional distribution of Y.pestis strains in Sichuan Province,and provided a scientific basis for prevention and treatment of plague.For determining the CRSIPR genotype,we cultivated and extracted nucleic acid from 132 Y.pestis strains which isolated from plague foci of Sichuan Province.Using PCR amplification,sequencing and analysis,all strains were genotyped with YPa,YPb and YPc primers of CRISPR.Results showed that we found 17 spacers,including 9 of YPa,5 of YPb and 3of YPc,and a newly kind of space array was discovered in 132 strains of Y.pestis.All pathogens were divided into 2 CRISPR gene clusters(Cc3′,Ca7)and 3 genotypes(37′,22 and sc01Type).Microtus fuscus strains were genotype 37′,and a strain isolated from plague patients in Shiqu county was genotype 22,and Marmota himalayana strains were genotype 22 in Dege County,and Marmota himalayana strains were genotype sc01 in Batang County,Litang County,Yajiang County and Xinlong County.Strains of Y.pestis in Sichuan Province have a strongly regional distribution characteristics.
