摘 要:目的:探知克里雅人线粒体DNA(mtDNA)遗传结构和遗传多样性,构建人群mtDNA遗传数据库。方法:运用二代测序技术对70个来自达里雅布依的克里雅人mtDNA基因组全序列进行了测序和分型。结果:70个个体共检出变异位点256处,控制区变异位点71处,编码区变异位点185,其中多态性位点245处。A263G、A750G、A4769G、A8860G、A15326G5个位点变异率为100%。界定了20种单倍型,高频率单倍型有F1a1(15.7%)、M8a2a1(12.9%)、H1t(11.4%)、M11a2(10.0%)。主成分分析结果表明:克里雅人与维吾尔族相聚的最近,与藏族和壮族相离较远,且中亚各人群相聚的较近。多态性指标显示,克里雅人π=0.1827、HD=0.9264、DP=0.9132,遗传多样性丰富。人群邻接系统进化树结果揭示,克里雅人与新疆维吾尔族人群亲缘关系最近,与藏族人群亲缘关系远。结论:克里雅人mtDNA遗传结构与维吾尔族人群最接近、亲缘关系最近,具有明显的亚欧混合现象。这与丝绸之路贸易的兴起昌盛衰落伴随着人群交流密不可分,是人群长期基因融合的结果。Objective:To explore the genetic structure and genetic diversity of Keriyan in Xinjiang,and to construct genetic database of mitochondrial DNA(mtDNA).Methods:Seventy mtDNAs of Keriyan from Daliyabuyi were sequenced and classified by the second generation sequencing technology.Results:The sequencing results showed that 256 variation sites were detected in 70 individuals including 71 sites in the control region and 185 sites in the coding region,which had 245 polymorphic locis.Among them,five variation rates were 100%such as A263G,A750G,A4769G,A8860G,and A15326G.Twenty haplotypes were detected with high-frequent appearance of F1a1(15.7%),M8a2a1(12.9%),H1t(11.4%)and M11a2(10.0%).The principal component analysis results showed that Keriyan and Uygur gathered together recently,which was far from Tibetan and Zhuang ethnicity,but closer to central Asian populations.The polymorphism index showed that nucleotide diversity was 0.1827,haplotype diversity was 0.9264,individual identification rate was as high as 0.9132 with rich genetic diversity.The population system phylogenetic tree revealed that Keriyan had closer genetic relationship with Xinjiang Uygur and far away from Tibetans.Conclusion:The analysis of genetic characteristics of keriyan from multiple perspectives shows the Eurasian mixing in the mtDNA genetic structure and its closest genetic relationship with Xinjiang Uygur people.This is the result of the long-term genetic integration due to the rise and decline of trade on the Silk Road.
