摘 要:为探究环境内分泌物对鱼类生长的毒性影响及作用机制,以鲤(Cyprinus carpio)为实验动物,利用三苯基氯化锡(TPT)为目标毒物,设定对照组和0. 024、0. 24、2. 4μg/L三个TPT暴露组,不同时间(7、21、48 d)暴露取样,通过检测血浆生长激素(GH)和胰岛素样生长因子(IGF-I)水平及肝脏生长激素受体(GHR)、IGF-I基因和脑垂体GH基因的表达量,探讨三苯基锡暴露对鲤GH/IGF-I轴毒性效应的影响及其机制。结果表明:暴露7 d时,高浓度的TPT(0. 24、2. 4μg/L)促进垂体GH的合成与分泌,而IGF-I的合成与分泌及肝脏IGF-I基因的表达受到显著抑制;暴露21、48 d时,血浆GH、IGF-I含量显著下降,脑垂体GH基因及肝脏GHR、IGF-I基因的表达均受到显著性抑制且呈剂量效应关系。研究结果表明,鲤血浆GH、IGF-I含量及肝脏GHR、IGF-I基因和垂体GH基因的表达量对不同浓度的TPT反应敏感,可以作为监测环境污染物对鱼类GH/IGF-I轴毒性影响的指示因子。Cyprinus carpio were exposed to 0,0.024,0.24,2.4μg/L Triphenyltin(TPT)for 7,21,48 days to countersign the effects of environmental endocrine disruptors on development of fish and explore the mechanisms.Hematological parameters and the expression of genes related GH/IGF-I axis were analyzed to probe into the toxic effects of TPT on the growth and GH/IGF-I axis of C.Carpio.The results showed that after 7 days exposure,high concentration TPT(0.24 and 2.4μg/L)irritated the synthesis and secretion of pituitary GH,while the synthesis and secretion of IGF-I and the expression of hepatic IGF-I were significantly inhibited.Plasma GH and IGF-I contents were significantly decreased,and the ex pression of pituitary GH and GHR and IGF-I in liver were significantly inhibited with dose-effect relationship after TPT(0.24 and 2.4μg/L)exposed for 21days and 48days.This study indicated the content of plasma GH and IGF-I as well as the expression of pituitary GH and GHR,IGF-I in liver was sensitive to different concentrations of TPT reaction.So these could be used as monitors to detect the toxicity effects of environmental pollutants on the growth and GH/IGF-I axis in fisheries.
