摘 要:目的:观察葛根素对心肌缺血再灌注损伤(MIRI)大鼠血清C反应蛋白(CRP)、一氧化氮(NO)及磷酸激酶(CK)含量的影响,探讨其抗心肌缺血再灌注损伤的作用机制。方法:建立IR模型,治疗组尾静脉注射葛根素注射液20mg/kg,记录结扎前5min、结扎或穿线后5min、再灌注5min、1h心电图,实验结束右心房采血测定各组血清中CRP、NO和CK的含量。结果:结扎大鼠冠状动脉左室支5min时各组心电图与假手术组相比,ST段抬高、T波高耸,治疗组ST段抬高程度低于假手术组和IR模型组;IR模型组血清CRP、NO和CK含量与假手术组比较差异显著(P <0.01);葛根素治疗组可以对抗CRP、CK升高和NO下降,与I/R模型组比较差异显著(P <0.05)。结论:葛根素能够减轻大鼠心肌缺血再灌注损伤,其可能机制与减少心肌细胞释放CRP,抑制炎症反应;升高NO,减少心肌酶漏出有关。Objective:To observe the effect of puerarin on the content of serum CRP,NO and CK in rats with MIRI,and explore active mechanism.Methods:The IR model was established.The treatment group was injected with puerarin injection 20mg/kg.The contents of serum CRP,NO and CK were record before ligation 5 minutes,after ligation or stringing 5 minutes,reperfusion 5 minutes and 1 hour ECG.Results:The ECG of left ventricular branch of coronary artery after 5 minutes in rats with ligation in each group was compared.ST segment and T wave in the other groups were higher than the sham operation group.Degree of ST segment elevation in the treatment group was lower than the sham operation group and IR model group.The difference in content of serum CRP,NO and CK levels betwwen the IR model group and the sham operation group were significantly(P<0.01).The puerarin can counteract the elevation of CRP and CK and drop of NO.Those show significant difference between the puerarin treatment group and IR model group(P<0.05).Conclusion:Puerarin can reduce myocardial ischemia reperfusion injury in rats.The mechanism may be related to reducing the release of CRP from cardiac myocytes,inhibiting inflammatory response,increasing no and reducing myocardial enzyme leakage.
