摘 要:DREB基因家族在调控植物生长发育和响应多种逆境胁迫中起着至关重要的作用。为了全面分析小麦中DREB基因家族成员的特性,为小麦DREB家族基因的发掘和利用提供参考,本研究采用生物信息学方法进行了全面的全基因组范围搜索,共获得了204个TaDREB基因。利用系统发育分析将其分为6个亚组,并进一步进行了染色体定位、基因结构和保守蛋白基序以及基因复制事件分析,系统研究了这些TaDREB基因的进化特征。此外,基于同源性分析的方法,构建了小麦DREB基因与其他基因之间的调控网络,鉴定到13个参与网络调控的TaDREB基因,并确定了179对互作关系。通过分析现有的RNA-seq数据,研究了TaDREB在不同组织和不同逆境胁迫下的表达情况,并鉴定到11个组织特异表达和多个响应逆境的基因。最后选取5个TaDREB基因,通过qRT-PCR技术验证了它们在热胁迫条件下的表达,结果证实这些基因均能对热胁迫产生响应。The DREB gene family is one of the most important gene families in plants,which plays the vital role in regulating plant growth and development as well as response to diverse stresses.Although DREBs have been thoroughly characterized in many plant species,the genome-wide identification of DREB gene family in wheat has not been reported yet.In order to comprehensively analyze the gene and protein properties of DREB gene family members in wheat,and provide reference for the discovery and utilization of wheat DREB family genes,in this study,a comprehensive genome-wide screening was performed to identify DREB gene family in T.aestivum and a total of 204 TaDREBs were obtained.Phylogenetic analysis classified them into six subgroups.Chromosomal localization,gene structure and conserved protein motif as well as gene duplication events analysis were further performed to systematically investigate the evolutionary features of these TaDREB genes.Furthermore,the regulatory network between TaDREB and other genes were constructed using the orthology-based method,and 13 TaDREBs were found to be involved in the regulatory network and 179 network branches were identified.The expression profiles of TaDREB in different tissues and under diverse stresses were investigated using the available RNA-seq data and 11 tissue-specific and several stress-responsive TaDREB genes were identified.Finally,five TaDREB genes were selected to validate their expression under heat stress treatments using qRT-PCR method and results verified that these TaDREBs could response to heat stress.
