钢鹅mtDNA全序列的克隆和生物信息学分析

摘  要:【目的】探究钢鹅的遗传多样性和系统进化的关系。【方法】参照NCBI上近缘物种线粒体基因组序列设计18对引物,运用DNA克隆和测序技术获得钢鹅mtDNA基因组序列,对其进行生物信息学分析。【结果】钢鹅mtDNA基因组序列总长16 739bp,其中含有22个tRNA基因、2个rRNA基因、13个蛋白质编码基因和1个非编码控制区域(D-loop区),基因组成及排列顺序与其他鸟类类似,A、T、C和G碱基含量分别为30.18%,22.54%,32.18%和15.10%。tRNAScan-SE Search Server在线软件分析表明,tRNA均是4个恒定臂的三叶草二级结构。对钢鹅mtDNA D-loop区序列的分析发现,其中含有LSP/HSP、ETAS1-2、Goose hairpin、E-box、F-box、D-box、C-box、CSB1-box及CSB-like保守框。用MEGA 6.0采用最大似然法基于D-loop区和线粒体全序列构建进化树,遗传进化分析表明,钢鹅与鸿雁、灰雁的亲缘关系最近。【结论】获得了钢鹅mtDNA全序列,进一步明确了其生物学信息。【Objective】The purpose of this paper was to investigate the genetic diversity and phylogenetic evolution of Gang goose.【Method】In this study,18 pairs of primers were designed with Primer 5.0,and the mitochondrial DNA(mtDNA)genome sequence of Gang goose was amplified by DNA sequencing technique,with reference to the mitochondrial genome sequences in the NCBI.【Result】The sequence of the mtDNA genome of Gang goose was 16 739 bp,which contained the 22 tRNA genes,the 2 rRNA genes,13 protein coding genes and one non-coding region(D-loop region).The contents of A,T,C and G were 30.18%,22.54%,32.18%and 15.10%,respectively.Online tRNAScan-SE Search Server analysis showed that tRNA had 4 constant arms liking a clover.The analysis of D-loop sequence showed that it contained LSP/HSP,ETAS1-2,Goose hairpin,E-box,F-box,D-box,C-box,CSB1-box and CSB-like.MEGA 6.0 with the maximum-likelihood algorithm and D-loop and mitochondrial phylogenetic tree showed that Gang goose,Anser cygnoides and Anser anser had close genetic relationship.【Conclusion】The whole sequence of mitochondrial genome of Gang goose was obtained and its biological information was clarified.
