摘 要:目的观察miR-125b在鼻息肉组织中的表达,并探讨其意义。方法收集30例鼻息肉样本和16例正常下鼻甲样本,采用实时定量PCR法检测miR-125b表达;转染miR-125b模拟物(miR-125b mimics)至鼻息肉上皮细胞,分别采用CCK-8试验和流式细胞术检测miR-125b表达对鼻息肉上皮细胞增殖和凋亡的影响,Western blotting试验检测细胞中PI3K/Akt信号通路相关蛋白表达。结果 miR-125b在鼻息肉中表达低于正常下鼻甲(P <0. 05);在鼻息肉上皮细胞中过表达miR-125b则细胞增殖受到抑制、细胞凋亡增加(P均<0. 05),同时,细胞中PI3KCD和p-Akt蛋白表达均下降(P均<0. 05)。结论 miR-125b在鼻息肉中表达降低,miR-125b可能通过负调控PI3K/Akt信号通路活性,促进鼻息肉上皮细胞增殖、抑制凋亡,进而诱导鼻息肉形成和发展。Objective To detect the expression of miR-125b in the nasal polyps and to explore its possible mechanism.Methods Thirty cases of nasal polyps and 16 cases of normal inferior turbinate were collected,and the expression of miR-125b was detected by real-time quantitative PCR.We transfected miR-125b mimics to epithelial cells of nasal polyps.CCK-8 assay and flow cytometry was used to detect the effects of the expression of miR-125b on proliferation and apoptosis of epithelial cells of nasal polyps.Western blotting was used to detect the expression of PI3K/Akt signaling pathway-related proteins.Results The expression of miR-125b in the nasal polyps was lower than that in the normal inferior turbinate(P<0.05);the overexpression of miR-125b in the epithelial cells of nasal polyps inhibited the cell proliferation and increased the apoptosis(both P<0.05).At the same time,the expression of PI3KCD and p-Akt protein decreased(both P<0.05).Conclusion The expression of miR-125b inthe nasal polyps decreases,and miR-125b may promote the proliferation and inhibit apoptosis of epithelial cells of nasal polyps by negatively regulating the activity of PI3K/Akt signaling pathway,and thereby induce the formation and development of nasal polyps.
