摘 要:为了分析lpfC基因对丙型副伤寒沙门菌生物学特性及致病性的影响,本研究采用Red同源重组系统构建丙型副伤寒沙门菌lpfC基因缺失株,并利用低拷贝质粒pSTV28构建其互补株。然后比较分析野生株、缺失株与互补株之间生长特性、生化特性、黏附侵袭能力、动物致病力等生物学特性差异。结果显示,lpfC基因的缺失不影响丙型副伤寒沙门菌的生长特性和生化特性,但缺失lpfC基因可导致丙型副伤寒沙门菌黏附侵袭细胞的能力及对小鼠致病力显著降低。本研究为进一步了解丙型副伤寒沙门菌lpfC基因的功能奠定了基础。To study the role of lpfC gene of Salmonella paratyphi C in biological functions and pathogenesis,the lpfC deletion mutant of Salmonella paratyphi C and its complementary strain were constructed using the Red recombinant system and low-copy plasmid pSTV28.Subsequently,the growth property,biochemical properties,adhesion and invasion capacity,and virulence in mice of this lpfC deletion mutant were analyzed and compared with the wild-type strain and complementary strain.The results showed that deletion of lpfC gene did not affect the growth property and biochemical properties.However,the adhesion and invasion capacity and virulence in mice significantly decreased in the lpfC deletion mutant.This study provided promising data for further research of the functions of the lpfC gene in Salmonella paratyphi C.
