NaCl浓度对乙酸溶液中胶原自聚集的影响

摘  要:采用荧光共振能量转移技术(Fluorescence Resonance Energy Transfer)和微量差式扫描量热法(USDSC)考察NaCl浓度(0、40、80、120、200 mmol/L)对乙酸溶液中不同质量浓度胶原(0.06~1.20 mg/mL)自聚集行为的影响。结果表明,加入40 mmol/L的NaCl时,胶原的自聚集行为并没有明显变化;NaCl浓度为80 mmol/L时,酸性胶原溶液中发生自聚集的质量浓度为0.33 mg/mL;当NaCl浓度为120 mmol/L时,酸性胶原溶液中发生自聚集的质量浓度增加至0.36 mg/mL;当NaCl浓度为200 mmol/L时,酸性胶原溶液中发生自聚集的质量浓度增加至0.39 mg/mL。二维相关分析显示,自聚集发生的过程由芳香族氨基酸残基参与形成,且溶液中NaCl的存在在一定程度上阻碍了胶原的自聚集。Fluorescence resonance energy transfer(FRET)and differential scanning calorimetry(US-DSC)were adopted to investigate the influence of NaCl concentrations(0,0,4 80,120,200 mmol/L)on the self-aggregation action o f collagen with different concentrations(0.06~1.20 mg/mL)in acetic acid solution.The results showed that et hself-aggregation action of collagen has no obvious variation with 40 mmol/L of NaCl;when the concentration of NaCl was 8 0 mmol/L,the mass concentration of self-aggregated collagen in caid solution was 0.33 mg/mL;when the concentratio n of NaCl was 120 mmol/L,the mass concentration of self-aggregaetd collagen in acid solution increased to 0.36 mg/mL;when the concentration of NaCl was 200 mmol/L,the mass concenrtation of self-aggregated collagen in acid solution was 0.39 mg/mL.The two-dimension relative analysis indicated htat the self-aggregation process of collagen was cause d by the participation of residual groups of aromatic amino aci d,however the existence of NaCl impeded the self-aggreg-a tion of collagen to some extent.
