摘  要:【目的】分析斜纹夜蛾普通气味结合蛋白(GOBP)基因(SlitGOBP2)的时空表达模式,为深入研究SlitGOBP2蛋白在斜纹夜蛾生长发育过程中的生理功能提供参考。【方法】以斜纹夜蛾为试验材料,分别收集并提取斜纹夜蛾卵、1~6龄幼虫、蛹共8个不同发育时期和雌、雄成虫触角、喙、胸、前足、中足、后足、腹部、翅膀共16个不同组织的RNA;以斜纹夜蛾Actin为内参基因,采用半定量RT-PCR分别对SlitGOBP2基因在雌、雄虫不同组织及不同发育阶段的基因表达模式进行比较分析。【结果】SlitGOBP2基因在斜纹夜蛾雌、雄虫的触角、喙、胸、前足、中足、后足、腹部和翅膀各组织中均有表达,其中在雌、雄虫触角中的表达水平均最高,在雌虫的喙、前足、后足和翅膀次之且明显高于雄虫的相应部位;雌、雄虫胸部和腹部的表达水平基本相同;而雌虫在中足的表达水平略低于雄虫。SlitGOBP2基因在斜纹夜蛾1~4龄幼虫和蛹后期均有表达,且在1龄幼虫和蛹后期中表达量相对较高,而在卵期、5和6龄幼虫中不表达。【结论】SlitGOBP2蛋白除具有取食、寻找寄主和产卵场所等基本功能外,雌虫可能还具有感受其他特殊信息素的功能。【Objective】The spatio-temporal expression patterns of odorant binding protein(GOBP)gene(SlitGOBP2)in Spodoptera litura were analyzed in this study,which provided scientific reference for further study of the physiological functions of SlitGOBP2 protein in the growth and development of S.litura.【Method】In this study,8 different developmental stages of S.litura including eggs,1st-6th instar larvae and pupa,and 16 different tissues including male and female adult antennae,proboscis,thorax,forefoot,middle foot,hind foot,abdomen and wings were collected and used to extract RNA of these tissues.On the basis of this,the gene expression pattern of SlitGOBP2 in different developmental stages and different tissues of female and male S.litura were compared by semi-quantitative RT-PCR with Actin as the internal reference gene.【Result】The tissue expression pattern of SlitGOBP2 indicated that the SlitGOBP2 was expressed in the antennae,proboscis,thorax,forefoot,middle foot,hind foot,abdomen and wing of male and female S.litura,and the expression level of antennae of male and female S.litura was higher than others.The expression in proboscis,forefoot,hind foot and wing of female ranked the second and they were higher than the corresponding tissues of male.The expression in thorax and abdomen of male and female was basically the same,but expression in middle foot of female was slightly lower than male.The gene SlitGOBP2 was expressed in the 1st-4th instar larva and pupa at late period of S.litura,but not in egg,5th and 6th instar larva,and the expression level in the1st instar larva and pupa at late period was high.【Conclusion】In addition to basic functions such as feeding,finding host and spawning sites,SlitGOBP2 protein may also have the ability to recognize and bind other specific pheromones from males.
