摘  要:目的观察右美托咪定(Dex)预处理对脾切除老年大鼠术后学习记忆能力的影响及机制。方法将健康雄性无特定病原体(SPF)级老年SD雄性大鼠60只按随机数字表法分为对照组、脾切除组和Dex处理组(Dex组)各20只。Dex组大鼠于脾切除手术前30 min经腹腔注射Dex 15μg/kg,对照组和脾切除组大鼠腹腔注射等量生理盐水。3组大鼠于术后7 d进行Morris水迷宫试验和旷场试验,行为学测试完毕后处死大鼠,检测其海马组织炎性因子IL-1、IL-6、CRP的浓度,海马淀粉样β蛋白、Tau-Thr181蛋白、磷酸化Tau-Ser356蛋白的表达,以及海马CA1区细胞凋亡数目,比较3组的差异。结果与对照组比较,脾切除组大鼠平均游泳距离均延长,探索原平台时间和探索次数缩短/减少(P均<0.05),旷场直立次数和跨格次数减少、中央区停留时间均缩短(P均<0.05);海马组织炎性因子IL-1、IL-6、CRP的浓度均增加(P均<0.05),海马组织淀粉样β蛋白,Tau-Thr181蛋白,磷酸化Tau-Ser356蛋白均增加(P均<0.05);凋亡阳性细胞数目增加(P<0.05)。与脾切除组比较,Dex组大鼠平均游泳距离均缩短,探索原平台时间和探索次数增加(P均<0.05);旷场直立次数和跨格次数、中央区停留时间均增加(P均<0.05);海马组织炎性因子IL-1、IL-6、CRP的浓度均减少(P均<0.05),海马组织淀粉样β蛋白,Tau-Thr181蛋白,磷酸化Tau-Ser356蛋白表达均降低(P均<0.05);Tunnel凋亡阳性细胞数目减少(P<0.05)。结论 Dex可改善脾切除老年大鼠术后学习记忆能力,其机制可能与其抑制海马炎性反应及淀粉样β蛋白、磷酸化Tau蛋白的表达,抑制海马神经元细胞的凋亡有关。Objective To observe the effect and mechanism of dexmedetomidine(Dex)pretreatment on the learning and memory capabilities in elderly rats after splenectomy.Method Sixty healthy SDFgrade aged male SD rats were randomly divided into the control(n=20),splenectomy(n=20)and dexmedetomidine pretreatment groups(Dex group,n=20)using the random number table.In the Dex group,intraperitoneal injection of dexmedetomidine at a dose of 15μg/kg was administered 30 min before splenectomy,and an equivalent quantity of normal saline was given in the control and splenectomy groups.At postoperative 7 days,the rats were subject to Morris water maze and open field test.The animals were sacrificed after behavioral tests.The hippocampal tissues were immediately removed for the determination of interleukin1(IL1)IL6,Creactive protein(CRP)using ELISA and the expression of amyloidβ,TauThr181 and phosphorylated TauSer356 proteins by Western blot.TUNEL method was utilized to detect the quantity of apoptotic cells in hippocampal CA1 area.Results Compared with the control group,the average swimming distance was significantly prolonged,the duration and frequency of exploring the original platform were considerably shortened,the number of standing and crossing and the residence time in the central district were significantly shortened in the open field(allP<0.05).Besides,the expression levels of IL1,IL6 and CRP in the hippocampal tissues were significantly upregulated(allP<0.05),and the expression levels of amyloidβ,TauThr181 and phosphorylated TauSer356 proteins were considerably upregulated and the quantity of apoptotic cells was significantly increased in the splenectomy group compared with the control group(allP<0.05).Compared with the splenectomy group,the average swimming distance was significantly shortened,whereas the duration and frequency of exploring the original platform were considerably increased,the frequency of standing and crossing in the open field was considerably increased the residence time in the central area was signific
