表达猪瘟病毒E2和猪繁殖与呼吸综合征病毒GP5蛋白重组腺病毒动物免疫保护试验

摘　　要：为研究共表达猪瘟病毒(CSFV)E2基因和猪繁殖与呼吸综合征病毒(PRRSV)GP5基因的重组腺病毒(rAd-E2-GP5)的免疫保护效果,将rAd-E2-GP5免疫34日龄~38日龄的健康断奶仔猪2次(间隔20d),同时设商品苗和空白对照组,二免21d后攻毒。ELISA结果显示,rAd-E2-GP5免疫后可检测到抗CSFV特异性血清抗体,未检测到抗PRRSV特异性血清抗体。组织核酸RT-PCR检测、临床症状和病理变化观察结果显示,重组腺病毒组分别攻击CSFV、PRRSV、CSFV+PRRSV强毒,死亡率为0,分别有0/5、4/5、4/5头表现临床症状,2/5、5/5、5/5头有眼观病变,0/5、4/5、4/5头检出PRRSV;商品苗组分别攻击CSFV、PRRSV、CSFV+PRRSV强毒,死亡率为0,分别有0/5、1/5、1/5头表现临床症状,3/5、4/5、3/5头有眼观病变,0/5、1/5、1/5头检出PRRSV;重组腺病毒组和商品苗对照组组织中均未检测到CSFV;空白对照组5/5头表现典型临床症状,5/5头死亡,5/5头检出PRRSV或CSFV。说明构建的rAd-E2-GP5能保护致死量的CSFV、PRRSV和CSFV+PRRSV强毒的攻击,但大部分猪有临床症状和病理变化。后续的研究中仍需要进一步加强重组疫苗对PRRS保护效果的试验,为CSF和PRRS联合疫苗的研发和实际应用奠定基础。In order to study the immune protective effects of recombinant adenovirus rAd-E2-GP5 co-expressing CSFV E2 gene and PRRSV GP5 gene,the healthy weaning piglets aged 34 to 38 days were immunized with rAd-E2-GP5 for twice(the interval was 20 days).Commercial vaccine and blank control groups were set up at the same time.The challenge was carried out 21-day after second vaccination.ELISA results showed that anti-CSFV specific serum antibody could be detected after rAd-E2-GP5 immunization,and no anti-PRRSV specific serum antibody was detected.The results of RT-PCR of nucleic acid of tissues,clinical symptoms and pathological lesions showed that the mortality was 0 after that the recombinant adenovirus vaccination challenged with CSFV,PRRSV and CSFV+PRRSV respectively.There were 0/5,4/5 and 4/5 having clinical symptoms,2/5,5/5 and 5/5 having grossly pathological lesions,and 0/5,4/5,and 4/5 being detected with PRRSV.No CSFV was detected in the tissues of the recombinant adenovirus group and commercial vaccine control group.In blank control group,5/5 showed typical clinical symptoms,5/5 were dead,and 5/5 were detected with PRRSV or CSFV.The results showed that the constructed rAd-E2-GP5 can protect the challenge of CSFV,PRRSV and CSFV+PRRSV with lethal dose,but most pigs had clinical symptoms and pathological lesions.The subsequent studies still need to further strengthen the protective effect of recombinant vaccine on PRRSV in order to lay the foundation for the R&D and practical application of the combined vaccines of CSFV and PRRSV.
