摘 要:目的观察兔自体骨髓间充质干细胞(BMSC)移植修复前交叉韧带损伤的效果及对兔血清尿激酶型纤溶酶原激活物(u-PA)、转化生长因子β(TGF-β)、白细胞介素1(IL-1)、IL-4水平的影响。方法将60只健康成年雄性新西兰大白兔随机分为3组,每组20只。模型组、移植组制作右前肢前交叉韧带损伤模型,于胫骨和股骨隧道内分别注入生物蛋白胶、BMSC和生物蛋白胶的混合物,正常组不做任何处理。术后2、4、6、8周,取静脉血检测血清u-PA、TGF-β、IL-1、IL-4,处死后测定前交叉韧带再次断裂所需拉力。结果术后2、4、6、8周时,移植组和模型组血清u-PA、TGF-β、IL-1、IL-4水平逐渐降低,前交叉韧带再次断裂所需拉力均逐渐增大,并向正常组水平趋近;术后同一时点,血清u-PA、TGF-β、IL-1、IL-4水平模型组>移植组>正常组(P均<0. 05),前交叉韧带再次断裂所需拉力模型组<移植组<正常组(P均<0. 05)。结论兔自体MSC移植可以降低血清u-PA、TGF-β、IL-1、IL-4水平,促进损伤部位的恢复。Objective To observe the effect of autologous bone mesenchymal stem cells(BMSC)transplantation on the repair of anterior cruciate ligament injury and its effects on the levels of Urokinase type plasminogen activator(u-PA),transforming growth factor-β(TGF-β),interleukin-1(IL-1),and interleukin-4(IL-4)in rabbits.Methods Sixty healthy adult male New Zealand white rabbits were randomly divided into three groups,with 20 in each.In the model group and the transplantation group,we made the models of right anterior cruciate ligament injury,and injected the mixture of biological protein glue,BMSC and biological protein glue into the tibia and femur tunnel,respectively;the normal group did not receive any treatment.At 2,4,6 and 8 weeks after surgery,the venous blood was taken to detect serum u-pa,TGF-β,and IL-4 concentrations.We measured the tension required for the re-fracture of the anterior cruciate ligament.Results At 2,4,6 and 8 weeks after surgery,the serum levels of u-PA,TGF-β,IL-1,and IL-4 of the transplantation group and the model group gradually decreased,and the tension required for the re-fracture of the anterior cruciate ligament gradually increased,and it was close to the level of the normal group.At the same time point after surgery,the serum levels of u-PA,TGF-β,IL-1,and IL-4 were in the following order:the model group>the transplantation group>the normal group(all P<0.05),and the tension required for the re-fracture of the anterior cruciate ligament:the model group<the transplantation group<the normal group(all P<0.05).Conclusion Autologous MSC transplantation can reduce serum levels of u-PA,TGF-β,IL-1 and IL-4,and promote the recovery of injured parts.
