摘 要:目的:探讨桦褐孔菌多糖(PIO)对哮喘气道重构的作用以及可能的作用机制。方法:40只雌性BALB/c小鼠随机分成对照组、模型组、桦褐孔菌多糖低剂量组(PIO-100)、桦褐孔菌多糖高剂量组(PIO-200)。体外卵清蛋白(OVA)诱导建立哮喘小鼠模型,支气管肺泡灌洗液(BALF)内细胞Diff-quik染色后观察细胞总数和各分类细胞数。肺组织切片HE染色和Masson染色观察肺部炎症情况。末次激发后检测小鼠气道高反应性,ELISA方法检测PIO对BALF中白细胞介素-4(IL-4)、IL-5、IL-13和MMP-9表达的影响。Western blot方法检测PI3K、Akt、NF-κB以及MMP-9表达水平的变化。结果:与对照组相比,模型组中炎症细胞计数、IL-4、IL-5和IL-13表达升高,p-PI3K、p-Akt和MMP-9的表达以及NF-κB p65核转位增高(P<0. 05); PIO干预后,炎症细胞数、IL-4、IL-5和IL-13的表达明显下降,p-PI3K、p-Akt和MMP-9的表达以及NF-κB p65核转位明显降低(P<0. 05)。结论:PIO通过调控PI3K/Akt/NF-κB/MMP-9信号通路发挥对哮喘小鼠气道重构的抑制作用。Objective:To investigate the effect of PIO on airway remodeling and the feasible mechanism.Methods:Forty female BALB/c mice were randomly divided into normal group,model group,PIO groups at low(PIO-100)and high(PIO-200)dose.Ovalbumin(OVA)was injected intraperitoneally and inhaled to induce the asthmatic model.The numbers of total cells and classified cells in bronchoalveolar lavage fluid(BALF)were observed by Diff-quik staining.The lung tissue inflammation was observed by HE staining and Masson staining and airway hyper reactivity(AHR)was measured in all of group.ELISA was used to detect the effect of PIO on the concentrations of interleukin-4(IL-4),IL-5,IL-13 and MMP-9 in BALF.Changes in PI3K,Akt,NF-κB and MMP-9 expression levels were detected by Western blot.Results:In the model group,the inflammatory cells,IL-4,IL-5 and IL-13 expressions increased,the expression levels of p-PI3K,p-Akt,MMP-9,and the nuclear translocation of NF-κB p65 increased comparing with the normal group(P<0.05);after PIO treatment,the number of inflammatory cells,IL-4,IL-5 and IL-13 expression were significantly decreased,p-PI3K,p-Akt and MMP-9 expression,and NF-κB p65 nuclear translocation were significant decreased(P<0.05).Conclusion:PIO can inhibit the airway remodeling in asthmatic mice by regulating PI3K/Akt/NF-κB/MMP-9 signaling pathway.
