摘 要:目的探讨脱氧核糖核酸酶(DNase)抑制肺炎链球菌(SP)青霉素耐药基因转移的作用。方法提取青霉素耐药SP(PRSP)的DNA,并进行相关试验鉴定;用PRSP的DNA转化SP标准菌株,转化后进行Optochin和苯唑西林药敏试验;用DNase抑制PRSP的DNA转化SP标准菌株,抑制转化后进行Optochin和苯唑西林药敏试验。结果提取DNA阶段,Optochin试验均显示阳性,苯唑西林药敏试验均显示青霉素不敏感,相关DNA检测均阳性;转化实验后,实验组和对照组的Optochin试验均显示阳性,实验组和对照组的苯唑西林药敏纸片抑菌环直径分别为(23.3±2.2)mm和(33.5±3.0)mm,两组比较差异有统计学意义(P<0.01);抑制转化实验后,实验组和对照组的Optochin试验均显示阳性,实验组和对照组的苯唑西林药敏纸片抑菌环直径分别为(29.6±4.7)mm和(17.6±9.4)mm,两组比较差异有统计学意义(P<0.05)。结论DNase具有抑制SP青霉素耐药基因转移的作用。Objective To explore the inhibition effect of Deoxyribonuclease(DNase)on penicillin resistance gene transfer among streptococcus pneumonia(SP).Methods The DNA of penicillin resistant SP(PRSP)was extracted and the related test of identification was conducted;SP standard strain was transformed by the DNA of PRSP.After the test of transformation,Optochin and Oxacillin susceptibility tests were conducted;the transfection was inhibited by DNase,Optochin and Oxacillin susceptibility tests were conducted after inhibition.Results In the DNA extraction stage,Optochin test was positive.Oxacillin susceptibility test showed that penicillin was not sensitive and the relevant DNA tests were positive.After transformation,Optochin test of the experimental group and the control group showed positive,the diameters of the bacteriostatic ring of Oxacillin-sensitive paper were(23.3±2.2)mm and(33.5±3.0)mm,respectively,the difference was statistically significant(P<0.01).After inhibition,Optochin test of the two groups were positive,the diameters of the bacteriostatic ring of the Oxacillin-sensitive paper in the experimental group and the control group were(29.6±4.7)mm and(17.6±9.4)mm,respectively,the difference was statistically significant(P<0.05).Conclusion DNase has the effect of inhibiting the gene transfer of penicillin resistance in SP.
