摘  要:2016年6月,四川眉山某养殖场爆发斑点叉尾急性传染病,发病1~3d即开始死亡,约7d达到死亡高峰,累计死亡率达60%,给养殖户造成了严重的经济损失。病鱼表现为离群独游,反应迟钝,食欲减退,发病后期出现"头穿孔"的现象。为明确该病的病原,本试验对病鱼进行了细菌学检测,使用16SrRNA PCR技术和微量生物化学鉴定方法进行了细菌的鉴定,并将分离的病原菌腹腔注射攻毒斑点叉尾,进行回归试验验证。试验结果显示,自斑点叉尾肾脏中分离到了一株革兰氏阴性短杆菌FW0606;经16SrRNA PCR测序,进化树分析显示,菌株FW0606与爱德华氏菌16SrRNA聚为一簇;进一步利用微量生化鉴定法确定该菌为爱德华氏菌。根据柯赫氏法则,健康斑点叉尾人工感染接种该菌后,试验鱼在12h开始出现死亡,72h内死亡率达到100%;病鱼表现出与自然发病相似症状,且同样分离到爱德华氏菌。为进一步对该病进行临床用药指导,对分离的爱德华氏菌进行了药敏试验鉴定,研究结果表明,爱德华氏菌FW0606对氟哌酸和氟苯尼考高度敏感,对丁胺卡那霉素、环丙沙星、阿奇霉素、卡那霉素和新霉素等中度敏感,对先锋霉素Ⅵ、氯洁霉素、四环素、美满霉素和强力霉素等耐药。An acute infectious disease of channel catfish Ictalurus punctatus was broken out in Meishan,Sichuan province in June 2016,resulting in death of most diseased fish within 1-3 days,cumulative mortality of 60%within one week.The diseased fish showed outbound swimming alone,slow response,loss of appetite,and the onset of“head piercing”phenomenon in late epidemic of the disease.A bacteriological test was carried out using 16S rRNA PCR,micro-biochemical identification and challenge test by intraperitoneal injection to identify the pathogen of the disease.The results showed that a strain of gram negative Brevibacterium named as FW0606 was isolated from the kidney.The 16S rRNA sequence revealed that the pathogen had the maximal similarity to Edwardsiella ictaluri and identified as Edwardsiella by micro-biochemical identification.According to Koch′s postulate,the same symptoms to those found in nature infection and death were observed in 12 h,with mortality rate of 100%72 h after the artificial infection.The antimicrobial susceptibility test showed that the strain was sensitive to norfloxacin and florfenicol,and resistant to vanguard ADMⅥ,clindamycin,tetracycline,minocycline,minocycline,and doxycycline.
