摘  要:目的筛选苏格木勒-3汤水提物中具有镇静催眠的成分。方法 102只大鼠随机分为空白组、SD模型组、苏格木勒-3汤水提物高及低剂量组、阳性组、白豆蔻-香旱芹水提物高及低剂量组、白豆蔻-荜茇水提物高及低剂量组、香旱芹-荜茇水提物高及低剂量组、白豆蔻水提物高及低剂量组、香旱芹水提物高及低剂量组、荜茇水提物高及低剂量组等17组,每组6只。除空白组外,其余各组大鼠腹腔射注300 mg/kg对氯苯丙胺酸混悬液制作大鼠失眠模型。造模成功后,空白组、模型组大鼠每天生理盐水灌胃,阳性组予阿普唑仑0. 12 mg/kg灌胃,苏格木勒-3汤水提物高及低剂量组分别予108、54 mg/kg的水提物灌胃,白豆蔻-香旱芹水提物高及低剂量组分别予84、42 mg/kg的水提物灌胃,白豆蔻-荜茇水提物高及低剂量组分别予82、41 mg/kg的水体物灌胃,香旱芹-荜茇水提物高及低剂量组分别予98、49 mg/kg的水提物灌胃、白豆蔻水提物高及低剂量组分别予33、16. 5 mg/kg的水提物灌胃、香旱芹水提物高及低剂量组分别予74、37 mg/kg的水提物灌胃、荜茇水提物高及低剂量组分别予29、14. 5 mg/kg的水提物灌胃,1次/d,连续灌胃7d。断头处死各组大鼠,取脑组织,ELISA法检测各组大鼠脑组织失眠指标5-羟色胺(5-HT)、神经递质γ-氨基丁酸(GABA)、白细胞介素1-β(IL-1β)及氧化还原指标超氧化物歧化酶(SOD)、丙二醛(MDA)。结果与空白组比较,模型组脑组织5-HT、GABA、SOD含量均降低,IL-1β、MDA含量升高(P均<0. 05)。与模型组比较,其余各组脑组织5-HT均升高(P均<0. 05);除苏格木勒-3汤水提物低剂量组外,其他组脑组织GABA含量均升高(P均<0. 05);苏格木勒-3汤水提物高、苏格木勒-3汤水提物低、白豆蔻-香旱芹水提物高、白豆蔻水提物低、荜茇水提物高组脑组织IL-1β降低(P均<0. 05);白豆蔻-香旱芹水提物高、白豆蔻-荜茇水提物低、香旱芹-Objective To screen out the sedative and hypnotic components of the Sugmel-3 decoction aqueous extract.Methods Totally 102 rats were randomly divided into 17 groups:the blank group,the SD model group,high-dose and low-dose Sugmel-3 decoction aqueous extract groups,positive group,and high-dose and low-dose Baidoukou-Xianghanqin aqueous extract groups,high-dose and low-dose Baidoukou-Biba aqueous extract groups,high-dose and low-dose Xianghanqin-Biba aqueous extract groups,high-dose and low-dose Baidoukou aqueous extract groups,high-dose and low-dose Xianghanqin aqueous extract groups,and high-dose and low-dose Biba aqueous extract groups,with six in each.In addition to the blank group,the rats in the other groups were injected intraperitoneally with 300 mg/kg p-chlorophenylalanine acid suspension to make models of insomnia.After successful modeling,the rats in the blank group and model group were given normal saline every day,the rats in the positive group were given alprazolam 0.12 mg/kg,those in the high-dose and low-dose Sugmel-3 decoction aqueous extract groups were given extract infusions of 108 and 54 mg/kg,and those in the high-dose and low-dose Baidoukou-Xianghanqin aqueous extract groups were given extract infusions of 84 and 42 mg/kg,those in the high-dose and low-dose Baidoukou-Biba aqueous extract groups were given extract infusions of 82 and 45 mg/kg,those in the high-dose and low-dose Xianghanqin-Biba aqueous extract groups were given extract infusions of 98and 49 mg/kg,those in the high-dose and low-dose Baidoukou aqueous extract groups were given extract infusions of 33 and 16.5 mg/kg,those in the high-dose and low-dose Xianghanqin aqueous extract groups were given extract infusions of 74 and 37 mg/kg,and those in the high-dose and low-dose Xianghanqin-Biba aqueous extract groups were given extract infusions of 29 and 14.5 mg/kg,1/d,for 7 d.The rats in each group were sacrificed and their brain tissues were taken.The insomnia index 5-hydroxytryptamine(5-HT),neurotransmitter alpha-aminobutyric acid
