摘 要:目的观察积雪草水煎液预先灌胃的阿尔兹海默病(AD)模型小鼠的学习记忆能力。方法 60只SPF级昆明小鼠,雌雄过半,均经Morris水迷宫筛选和训练,随机分为正常对照组、模型组、高剂量组、中剂量组、低剂量组及阳性药组。正常对照组、模型对照组以蒸馏水灌胃;高、中、低剂量组分别予0. 624、0. 312、0. 156 g/m L的积雪草水煎液灌胃,25 m L/kg;各组均干预30 d,第31天给药后30 min,除正常对照组外其余组小鼠腹腔注射2mg/kg的东莨菪碱制作AD模型。造模30 min时采用Morris水迷宫正式实验观察比较各组学习记忆能力(潜伏期、经过平台次数)。脱颈椎处死各组小鼠,取脑组织,用紫外分光光度计测定各组小鼠脑组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,采用全自动酶免分析系统分测定各组小鼠脑组织丙二醛(MDA)、乙酰胆碱酯酶(ACh E)、胆碱乙酰基转移酶(Ch AT)含量。结果与模型对照组比较,高、中、低剂量组小鼠潜伏期缩短、经过平台次数增加(P均<0. 05),且呈剂量依赖性;与正常对照组比较,模型对照组组小鼠潜伏期延长、经过平台次数降低(P均<0. 05)。与正常对照组比较,模型对照组小鼠脑组织内SOD、GSH-Px活性降低,MDA和ACh E含量增加,Ch AT含量减少(P均<0. 05)。与模型对照组比较,高剂量组小鼠脑组织GSH-Px活性升高,MDA含量降低,Ch AT含量升高(P均<0. 05)。结论积雪草水煎液预先灌胃可减轻AD模型小鼠学习记忆能力的减退,其机制可能为增加脑组织内Ch AT含量、提高GSH-Px活性,降低MDA含量。Objective To observe the learning and memory abilities in mice with Alzheimer's disease(AD)after pre-intragastric infusion of Centella asiatica water decoction.Methods Sixty SPF Kunming mice(half male and half female)were screened and trained by the Morris water maze before the formal experiment began.Then they were randomly divided into six groups:the control group,the model group,high-dose group,medium-dose group,low-dose group,and the positive drug group.Each group was administered relative drugs or water with the volume of 25 mL/kg by gavage for 30 days.The control group and the model group were administered distilled water;the high-,medium-,and low-dose groups were given 0.624,0.312 and 0.156 g/mL Centella asiatica water decoction,respectively.On the 31st day,each group(except the control group)was intraperitoneally injected with hyoscine to make the AD models.The learning and memory abilities of mice were tested by using the Morris water maze 30 minutes later.After that,all the mice were euthanized and their brains were taken out so that the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)could be measured by ultraviolet spectrophotometer,and the levels of MDA,acetylcholin esterase and choline acetyltransferase(ChAT)were detected by microlab FAME system.Results Compared with the model group,the latent period in the high-dose group,medium-dose group,and low-dose group was shorter and the number of mice passing the platform increased(P<0.05),with a dose-dependent manner.Compared with the control group,the latent period in the model group was prolonged and the number of mice passing the platform decreased(P<0.05).Compared with the control group,the activity of SOD,GSH-Px and the level of ChAT in the model group decreased,while the levels of MDA and AChE increases(P<0.05).Compared with the model group,the activity of GSH-Px and the level of ChAT in the high-dose increased,while the level of MDA decreased(P<0.05).Conclusion Pre-intragastric infusion of Centella asiatica water decoction ca
