摘 要:为明确甘肃省紫花苜蓿(Medicago sativa)种子携带的主要病毒病原,利用双抗体夹心酶联免疫吸附分析(DASELISA)方法对收集的甘肃省紫花苜蓿种子样品进行检测,并采用RT-PCR方法对DAS-ELISA检测呈阳性的代表性样本进行复检,结果表明,在52份紫花苜蓿种子样品中有45份携带苜蓿花叶病毒(alfalfa mosaic virus,AMV)、3份携带菜豆黄花叶病毒(bean yellow mosaic virus,BYMV),携带AMV和BYMV的样品分别占样品总数的86.5%和5.8%。该地区紫花苜蓿种子携带的病毒为AMV、BYMV。本研究为防治苜蓿病毒病提供了参考。The purpose of this study was to identify the primary viruses in alfalfa seeds collected from Gansu Province.Seeds were analyzed using a double-antibody sandwich enzyme-linked immunosorbent assay(DAS-ELISA)to detect the presence of alfalfa viruses.Several samples showing positive ELISA results were analyzed by reverse transcription-polymerase chain reaction(RT-PCR)and the coat protein-coding region of the detected viruses was sequenced and compared with the corresponding sequences available in databases.Among a total of 52 seed lots collected,Alfalfa mosaic virus(AMV)was detected in 45 samples and bean yellow mosaic virus(BYMV)was detected in three samples.The average rate of infection with AMV and BYMV was 86.5%and 5.8%,respectively.We conclude that the main viruses in alfalfa seed lots collected from Gansu Province are AMV and BYMV.
