中药复方多糖对不同MHC B-LβⅡ基因型鸡淋巴细胞TLR 4介导的TRIF依赖性途径的影响  被引量:4

摘  要:探讨中药复方多糖在不同MHC B-LβⅡ基因型鸡淋巴细胞中对TLR4信号转导通路下游TRIF依赖性途径主要元件的影响。采用PCR-SSCP方法将500羽京红1号蛋鸡按不同MHC B-LβⅡ基因型分组,采集不同MHC B-LβⅡ基因型鸡的外周血淋巴细胞,分别加入终质量浓度为100、75、50、0μg/mL(高、中、低、对照组)中药复方多糖共培养16、24、32、48h,收集细胞,采用Real-time PCR方法检测TRIF、IRF3、IFN-β基因mRNA的表达。结果显示:(1)与对照组相比,中药复方多糖能显著增强不同MHC B-LβⅡ基因型鸡TRIF、IRF3、IFN-βmRNA的表达量(P<0.05)。(2)中、低剂量中药复方多糖组AA基因型鸡TRIF、IRF3基因mRNA的表达量各个时间段均高于高剂量组,IFN-β在24、32、48h高于高剂量组,高剂量中药复方多糖组BB基因型TRIF、IRF3基因的表达量各个时间段高于其他剂量组,IFN-β在24、32、48h高于其他剂量组,低剂量中药复方多糖组中BC基因型TRIF、IRF3基因的表达量各个时间段高于其他剂量组,IFN-β在24、32、48h高于其他剂量组。表明中药复方多糖可能与淋巴细胞表面TLR4结合,可以通过下游TRIF依赖性信号转导通路,调控细胞免疫,并且对促进其信号通路主要元件mRNA的表达的最适cCHMPS剂量有一定的影响。To investigate effects of different doses of traditional Chinese medicine compound polysaccharides on TLR4 and dependent signal transduction pathway components of downstream TRIF in chicken lymphocytes of different MHC B-L and betaⅡgenotypes,PCR-SSCP technique was applied to group layer according to different MHC B-LβⅡgenotype,we collected the different MHC B-LβⅡgenotype from layer,isolated peripheral blood lymphocytes,added to cCHMPS which the final concentration was 100,75,50,0μg/mL co-cultured 16,24,32,48 h,and then we detected the expression of lymphocyte TRIF,IRF3,IFN-βmRNA using Real-time PCR method.The results showed that:(1)Polysaccharide extraction at different doesfrom traditional Chinese medicine could improve the chicken lymphocyte of TRIF,IRF3,IFN-βmRNA expression levels compared with the control group in each MHC B LβⅡgenotype chickens.(2)In high does of traditional Chinese medicinal polysaccharide extraction,AA genotype chicken lymphocyte of the TRIF,IRF3,IFN-βmRNA was lower than that of other groups,in high does of polysaccharide extraction from traditional Chinese medicinal,BB genotype chicken lymphocyte of the TRIF,IRF3,IFN-βmRNA was higher than that of other groups,in low does of polysaccharide extraction from traditioanl Chinese medicinal,BC genotype chicken lymphocyte of the TRIF,MYD88,IFN-βmRNA was higher than that of other groups.There results indicated that the mechanism of Chinese herbal compound polysaccharides played a role in regulating the immune in compounding Chinese medicine polysaccharide and cell surface TLR4 binding,activating the TRIF dependent signal transduction pathways,regulating cell immunity,and the traditional Chinese medicinal polysaccharide extraction optimum immunomodulatory does were different in each MHC B-LβⅡgenotype chickens.
