摘 要:目的研究钩吻素己在大鼠体内的毒物动力学、组织分布和排泄情况,为钩吻中毒案件中法医学鉴定的取材和毒理学研究的开展提供参考和依据。方法采用单次灌胃染毒法建立大鼠急性中毒模型,雄性和雌性大鼠分别灌胃给予低、中、高剂量的钩吻素己,于不同时间点采集血液样品;取大鼠于灌胃后5、15、60 min处死并采集心、肺、脾等组织;另取大鼠灌胃后收集24 h的尿液和粪便。应用液相色谱-质谱联用法(LC-MS/MS)测定各生物检材中钩吻素己的含量。结果钩吻素己经消化道吸收迅速,在小肠、胃、肝、胰中含量较高,在给药后24 h的尿液和粪便中原型药物排泄量很低。结论钩吻素己灌胃给药后分布快速而广泛,肝和胰可作为此类中毒案件的首选检材。另外,钩吻素己的毒性作用在大鼠体内存在性别差异。Objective To explore the toxicokinetics and tissue distribution of gelsenicine in rats after a single dosage,and to provide reference for the sampling in forensic practice and toxicology researches.Method Low,medium and high doses of gelsenicine were administrated intragastrically to different groups of male and female rats.The blood samples were collected at different time intervals.Rats were killed in batches after 5,15,60 minutes of the oral administration,and tissues such as heart,lung and spleen were collected.The urine and dung samples were collected within 24 hours after the oral administration.The concentrations of gelsenicine in different biological samples were determined by liquid chromatography-tandem mass spectrometry.Results The gastrointestinal absorption of gelsenicine was very fast.Gelsenicine’s concentrations in the intestine,stomach,liver and pancreas samples were higher than those in the other tissues.The amount of gelsenicine in the urine and dung samples was little.Conclusion The distribution of gelsenicine in rats was fast and wide.There was significant difference of the distribution between male and female rats.Besides,liver and pancreas could be used as the preferred biological samples for the identification of gelsenicine poisoning.
