摘 要:为揭示盐芥和拟南芥表皮蜡质中响应低温胁迫的关键化学组分及关键代谢基因表达的变化,以盐芥和拟南芥为材料,通过化学分析和荧光定量PCR技术,比较和研究了4℃低温胁迫处理下2种植物表皮蜡质的组成及蜡质代谢相关基因的表达差异。结果表明,与对照(22℃)相比,在4℃胁迫条件下,盐芥和拟南芥叶片表皮总蜡质含量以及蜡质各组成成分的含量均增加。经低温胁迫处理后,2种植物中组成蜡质的优势组分均未发生变化,表现为烷烃含量最多、初级醇含量次之、脂肪酸含量位居第3。对所选取的28个蜡质代谢相关基因的表达量进行分析,结果显示,盐芥中有23个基因上调,拟南芥中有13个基因上调,暗示盐芥和拟南芥中表皮蜡质代谢基因对低温的响应不同。The wax composition and related gene expression in the leaf epidermis of Thellungiella salsuginea and Arabidopsis thaliana under 4℃cold stress treatment(4℃)were compared and studied by chemical analysis and quantitative(real-time)reverse transcription-PCR design to reveal the key chemical composition and relative gene expression of epidermal wax responded to cold stress.The results showed that,in both species,after cold treatment the amounts of total cuticular wax and all wax components constituents on the leaf epidermis after cold treatment were higher than those in the control group(22℃).After cold stresstreatment,the dominant components of the wax composition did not change,with the alkane as the major class,followed by primary alcohol and fatty acidacid.Among 28 wax-associated genes,23 genegenes were up-regulated in T.salsuginea and 13 genes were up-regulated in A.thaliana under cold stress respectively.It suggested that T.salsuginea and A.thaliana responded differently to cold treatment concerning epidermal wax metabolism remained difference in response to cold stress between T.salsuginea and A.thaliana.
