溶藻弧菌TolB蛋白的生物信息学分析

摘　　要：对溶藻弧菌转运蛋白(TolB)进行结构和功能进行分析,设计其表位多肽重组疫苗,以提高溶藻弧菌TolB蛋白的免疫效果,运用生物信息学方法对溶藻弧菌TolB蛋白的系统进化关系、理化性质、高级结构和细胞抗原表位进行分析。进化关系结果显示,溶藻弧菌的TolB蛋白与副溶血性弧菌、霍乱弧菌和费氏弧菌的TolB蛋白具有较高同源性,溶藻弧菌TolB蛋白可能对这4种菌株的感染起到交叉免疫保护作用。溶藻弧菌TolB蛋白为亲水性蛋白,1—22位氨基酸为其信号肽位置,无跨膜结构并定位于细胞膜外;溶藻弧菌TolB蛋白二级结构中以无规则卷曲(40.00%)、α-螺旋(18.67%)和β-转角(11.33%)为主。溶藻弧菌TolB蛋白可能存在4个优势B细胞抗原表位,1个细胞毒性T淋巴细胞(CTL)抗原表位,1个辅助T细胞(Th)抗原表位,并重组拼接获得抗原性较好的TolB蛋白表位多肽。In order to analyze the structure and function of Vibrio alginolyticus transporter TolB protein and design its epigenetic polypeptide recombinant vaccine,further improving the immune effect of TolB protein,the phylogenetic relationships,physicochemical properties,advanced structure and cell epitope of TolB protein was analyzed by bioinformatics.The results of evolutionary relationship showed that TolB protein of Vibrio alginolyticus,Vibrio parahaemolyticus,Vibrio cholerae and Vibrio cholerae had high homology.TolB protein might play cross immune protection against the infection of these four strains.TolB protein was hydrophilic protein.And 1 to 22 amino acids of TolB were the signal peptide position,did not exist transmembrane structure and located outside the cell membrane.The secondary structure of the TolB protein mainly included random coil(40.00%),alpha helix(18.67%)and beta turn(11.33%).TolB protein had four B cell epitopes,one CTL cell epitope and one Th cell epitope,respectively.The TolB protein epitope polypeptides with better antigenicity was obtained by reorganization and splicing.
