摘  要:目的探讨葛根素对去势雌性大鼠骨质疏松症及PI3KAKT信号转导通路的影响。方法 40只大鼠随机平均分为假手术组、模型组、葛根素低剂量组和葛根素高剂量组,手术摘除大鼠卵巢以复制骨质疏松症大鼠模型。比较4组大鼠一般状态、股骨头病理切片和股骨头骨密度、血清中钙、磷、血管内皮生长因子(VEGF)和一氧化氮(NO)水平、股骨头中Akt和p-Akt蛋白表达水平。结果葛根素低剂量组和葛根素高剂量组大鼠给药后活动程度显著提高,毛发有色泽,精神状态良好;葛根素低剂量组和高剂量组股骨头病理较模型组显著改善;葛根素低剂量组和葛根素高剂量组大鼠股骨头骨密度、血清中钙和磷水平显著大于模型组,且葛根素高剂量组组大鼠的股骨头骨密度、血清中钙和磷水平、血清中VEGF和NO水平显著高于葛根素低剂量组(P <0. 05); 4组大鼠股骨头中Akt蛋白表达水平差异无统计学意义(P> 0. 05);葛根素低剂量组和葛根素高剂量组大鼠p-Akt蛋白表达水平显著高于模型组,且葛根素高剂量组大鼠p-Akt蛋白表达水平显著性高于葛根素低剂量组(P <0. 05)。结论葛根素对去势雌性大鼠骨质疏松症有治疗作用,作用机制可能为升高血清中促进血管新生活性因子(如VEGF和NO)水平的升高,进而促进血管新生,重建供血和血液循环,促进坏死部位新骨生成。Objective To investigate the effects of the puerarin on osteoporosis and 3K/AKT signaling pathway in ovariectomized female rats.Methods A total of 40 rats were randomly divided into sham operation group,model group,puerarin low-dose group and puerarin high-dose group.A rat model of osteoporosis was established by surgical removal of the ovaries of female rats.The general state,the pathological section and bone density of femoral head,serum levels of calcium,phosphorus,vascular endothelial growth factor(VEGF)and nitric oxide(NO),and the expression levels of Akt and p-Akt protein in femoral head were observed and compared.Results The activities of the rats in puerarin low-dose group and puerarin high-dose group were increased significantly after drug administration,the hair color and spirit of rats were in better condition.The pathological sections of femoral head in puerarin low-dose group and puerarin high-dose group were significantly improved,as compared with those in model group.The bone mineral density,serum levels of calcium,and phosphorus in puerarin low-dose group and puerarin high-dose group were significantly higher than those in model group,moreover the bone mineral denaity,serum levels of calcium,phosphorus,VEGF and NO in puerarin high-dose group were significantly higher than those in puerarin low-dose group(P<0.05).There were no significant differences in the expression levels of Akt protein in the femoral head among the four groups(P>0.05).The expression levels of p-Akt in puerarin low-dose group and puerarin high-dose group were significantly higher than those in model group.In addition p-Akt protein expression levels in puerarin high-dose group were significantly higher than those in puerarin low-dose group(P<0.05).Conclusion Puerarin has therapeutic effect on osteoporosis in ovariectomized female rats,and its action mechanism may be correlated with increasing the serum levels of new vascular active factors including VEGF and NO,promoting angiogenesis,reconstructing blood-supply and blood circulat
