摘  要:目的模拟西方人群高果糖含量的饮食结构及习惯,本研究以单纯高果糖饲料喂养的方式,尝试构建代谢综合征大鼠模型。方法选取7周龄健康无特定病原体雄性Wistar大鼠40只,根据体质量随机分为对照组和实验组,每组20只。实验组给予高果糖饲料喂养,对照组给予普通饲料喂养。动态监测大鼠体质量、腹围、血压等一般状况;6周后检测血糖和血脂相关指标,包括空腹血糖、胰岛素、TG、HDL-C、LDL-C和TC;分析胰岛素敏感指数(ISI)和胰岛素抵抗指数(IRI);测量内脏脂肪质量,并计算Lee's指数。同时采用多道生物记录仪测量左心室收缩压、左心室内压等。结果实验组大鼠内脏脂肪质量和内脏脂肪质量/体质量较对照组明显升高[(35.60±3.17)g vs(22.26±7.43)g,(7.26±2.23)%vs(5.16±1.20)%,P<0.05]。实验组大鼠空腹血糖、TG、TC、LDL-C、胰岛素、IRI、左心室收缩压和左心室舒张末期压明显高于对照组,HDL-C和ISI明显低于对照组,差异有统计学意义(P<0.05)。结论采用Wistar大鼠单纯高果糖饲料喂养6周,能够成功建立代谢综合征大鼠模型。Objective To establish a metabolic syndrome(MS)model of rats fed on simple high fructose diet in order to simulate the structure and habits of high fructose diets in western populations.Methods Forty healthy male Wistar rats aged 7 weeks without specific pathogens were randomly divided into control group(n=20)and experimental group(n=20)according to their body weight.Six weeks after the animals in experimental group were fed on high fructose diet and those in control group were fed on common chow,their body weight,abdominal circumference and blood pressure were recorded,their FPG,blood lipid and serum insulin,TG,HDL-C,LDL-C,TC levels were measured,their ISI and IRI were analyzed,their visceral fat mass was measured and Lee’s index was calculated.The Left ventricular systolic pressure and left ventricular pressure were measured with a multichannel biometric recorder.Results The visceral fat mass and visceral fat mass/BMI ratio were significantly higher in experimental group than in control group(35.60±3.17 g vs 22.26±7.43 g,7.26%±2.23%vs5.16%±1.20%,P<0.05).The FPG,serum TG,TC,LDL-C,insulin levels,IRI,left ventricular systolic pressure and left ventricular end diastolic pressure were significantly higher while the serum HDL-C level and ISI were significantly lower in experimental group than in control group(P<0.05).Conclusion The MS model of rats is successfully established by feeding the rats on simple high fructose diet for 6 weeks.
