摘 要:目的:探讨变异链球菌ldh-luc基因荧光报告株(荧光报告株)对氯己定的敏感性。方法:将变异链球菌荧光报告株S.mutans ldh-luc和野生菌株分别以浮离态和生物膜态培养于BHI,菌落计数法(CFU)检测氯己定对2种菌株的最小抑制浓度(MIC)、RLU、A_(600),以观测其生长状态及荧光活性的变化及相互关系。结果:氯己定对S. mutans ldh-luc基因荧光报告株和野生株的MICs均为(0. 25±0. 39)μg/ml。氯己定作用30 min后,浮游态和24 h生物膜S. mutans ldh-luc基因荧光报告株的RLU、A_(600)、CFU呈下降趋势;作用60 min后,RLU、CFU下降至检测水平以下,去除氯己定,加入BHI培养基培养90 min,仍未检出RLU,氯己定作用前后的差异具有统计学意义(P <0. 05)。结论:S. mutans ldh-luc基因荧光报告株能准确反映氯己定对生物膜中细菌的作用。Objective:To investigate the response of ldh-luc reporter strain of Streptococcous mutans(S.mutans)to chlorhexidine.Methods:MICs,RLU,A 600 and CFU of the wild type and ldh-luc reporter strain of S.mutans culture with BHI in a planutonic form and in biofilm form respectively to chlorhexidine were examined.Results:The MICs of chlorhexidine to both strains of S.mutans was(0.25±0.39)μg/ml.The sensitivities and response of the reporter strain and wild type to chlorhexidine were similarly,no matter in a planktonic culture or in biofilms.After 30 min treated by chlorhexidine,all of RLU,A 600 and CFU of ldh-luc reporter strain of S.mutans were decreased.After 60 min,none of RLU and CFU of ldh-luc reporter strain of S.mutans was detected.Conclusion:ldh-luc reporter strain of S.mutans may actually show the response of S.mutans to chlorhexidine.
