摘　　要：目的观察人正常肝细胞L-02上清液对转化生长因子(TGF)-β1刺激的体外培养人肝星状细胞株(HSC)-LX-2α-平滑肌肌动蛋白(α-SMA) mRNA和蛋白表达的影响。方法体外培养HSC-LX-2、L-02,设立HSC-LX-2正常组、模型组、L-02细胞上清原液组、1/2上清液组、1/4上清液组,分别孵育72 h,噻唑蓝染色法(MTT)检测HSC-LX-2细胞的抑制率,Western blot法检测各组α-SMA的表达,逆转录聚合酶链反应(RT-PCR)法检测各组α-SMA mRNA的表达。结果L-02细胞上清原液组、1/2上清液组α-SMA mRNA及α-SMA蛋白的表达有显著的抑制作用(P <0. 01,P <0. 05)。结论 L-02上清液能抑制HSC-LX-2细胞的活化和增殖。Objective To observe the effect of human normal liver cell(LO2)cells’supernatant liquid on the mRNA and protin expressions ofα-smooth muscle actin(α-SMA)in human hepatic stellate cell(HSC-LX-2)cells stimulated by transforming growth factor-β1(TGF-β1)in vitro.Methods HSC-LX2 cells and L-02 cells were cultured in vitro and divided into the blank group,model group,original L-02 cells’supernatant group,1/2 supernatant group and 1/4 supernatant group.After being incubated with each concentration of the L-02 cells’supernatant liquid for 72 h,the mRNA expression ofα-SMA was assayed by RT-PCR and the protein expression ofα-SMA was assayed by Western blot.Results There was a significant inhibitory effect on the expression ofα-SMA mRNA andα-SMA protein on the original L-02 cells’supernatant group and the 1/2 supernatant group(P<0.01,P<0.05).Conclusion LO2 cells’supernatant liquid can inhibit the activation and proliferation of HSC-LX-2 cells.
