摘  要:目的通过研究大鼠胰腺组织和胰岛β细胞系INS-1(大鼠源)和HIT-T15(仓鼠源)细胞中多巴胺(dopamine,DA)受体的分布以及INS-1细胞系的内分泌功能,为DA调节胰岛β细胞功能研究结果不一致的可能原因提供一定的实验依据。方法应用双标记免疫荧光染色方法观察在大鼠胰腺组织和胰岛β细胞系中DA受体分布的异同,以及胰岛素和胰高血糖素免疫荧光双标记结果的差异。应用放射免疫法检测INS-1细胞孵育液中胰岛素及胰高血糖素的浓度,观察细胞系的内分泌功能的改变。结果在大鼠胰腺组织中,DA受体D1、D2和D5分别表达在β细胞、δ细胞、α细胞和PP细胞上,β细胞只表达D1受体。然而在两种胰岛β细胞系INS-1和HIT-T15细胞上不仅有D1受体,还有D2和D5受体的分布。两种胰岛β细胞系均同时表现胰岛素和胰高血糖素免疫阳性,而在原位胰岛中分布于不同的细胞,进一步检测显示INS-1细胞系的孵育液中不仅含有胰岛素,也含有胰高血糖素。结论胰岛β细胞系DA受体分布及内分泌功能不同于原位β细胞。Objective To investigate distribution of dopamine(DA)receptors in rat pancreatic tissue andβ-cells lines(INS-1and HIT-T15cells)and further the endocrine function of INS-1cell.Methods Double-labeled immunofluorescence staining was used to observe the differential distribution of dopamine receptors and immunofluorescence of insulin and glucagon between in rat pancreatic tissue and tumorβ-cell lines.The insulin and glucagon content in the supernatant of INS-1cell line was detected by radioimmunoassay to observe the changes of endocrine function.Results In rat pancreatic tissue,DA receptors D1,D2,and D5were distributed in theβ-cells,δ-cells,andα-cells,PP-cells,respectively,that is,β-cells expressed only D1receptors.However,not only D1,but also D2and D5were distributed in the INS-1and HIT-T15β-cell lines.Insulin and glucagon immunoreactivities were simultaneously expressed in theβ-cell lines.Furthermore,both the insulin and glucagon were detected in the supernatant of INS-1β-cell line.Conclusion The present study for the first time demonstrated that the distribution of DA receptors and endocrine function inβ-cell lines are different from that in situ.
