摘 要:目的:探究皂荚中活性成分皂荚素对高胆固醇模型大鼠抗氧化能力和DNA损伤的影响,对大鼠血浆胆固醇以及其相关血脂生化指标的影响。方法:将48只健康大鼠,随机分为三个组:对照组、低浓度皂荚素组、高浓度皂荚素组。各组饲料皂荚素含量分别为0、80、200(mg/kg)。制作由不同浓度H2O2(0、6、12、18μmol/L)诱导的DNA损伤兼高胆固醇大鼠模型。实验期限为60d。实验结束,采血,应用试剂盒检测血清丙二醛(MDA)水平和谷胱甘肽过氧化物酶(GSH)活性,并采用单细胞凝胶电泳法测定淋巴细胞DNA的损伤情况。结果:与对照组比较,低浓度皂荚素组、高浓度皂荚素组MDA含量和血清GSH活性差异有统计学意义。低浓度皂荚素组和高浓度皂荚素组大鼠血清总胆固醇(TC)、甘油三酯(TG)、载脂蛋白B(apo B)的含量都有明显的降低,尤其以高浓度皂荚素组最为显著,同时,两组大鼠的胆汁酸排泄量增加,载脂蛋白AⅠ(apo A-Ⅰ)、高密度脂蛋白—胆固醇(HDL-C)水平影响不明显,差异无统计学意义。结论:皂荚素可有效降低血浆胆固醇水平。对于健康幼年大鼠血清GSH及脂质过氧化产物MDA含量有显著影响,对H2O2诱导的DNA损伤也表现出明显的保护作用。Objective:To investigate the effects of active component of Gleditsia sinensis sinensis on antioxidation and DNA damage in hypercholesterolemic rats,and the effect of Gleditsia sinensis on plasma cholesterol and blood lipid biochemical indexes in rats.Methods:48healthy rats were randomly divided into three groups:control group,low concentration Gleditsia sinensis group and high concentration Gleditsia sinensis group.The contents of Gleditsia sinensis sinensis were0,80and200(mg/kg)respectively.The DNA damage and high cholesterol rat model induced by H2O2(0,6,12,18μmol/L)of different concentrations was made.The experimental period was60days.The serum malondialdehyde(MDA)and glutathione peroxidase(GSH)activity were detected with kit,and the damage of lymphocyte DNA was detected by single cell gel electrophoresis.Results:Compared with the control group,the content of MDA and the activity of GSH in low concentration Gleditsia sinensis group and high concentration of Gleditsia sinensis sinensis were significantly different.Low concentration of Gleditsia sinensis sinensis and high concentration of Gleditsia sinensis sinensis serum total cholesterol(TC),triglyceride(TG),apolipoprotein B(apoB)content were significantly reduced,especially in high concentration of Gleditsia sinensis group,at the same time,the bile acid excretion in two groups of rats increased,apolipoprotein A-I(apoA-I),high density lipoprotein cholesterol(HDL-C)level effect is not obvious,no statistical significance.Conclusion:Gleditsia sinensis sinensis can effectively reduce the level of plasma cholesterol.It had significant effect on the content of GSH and MDA in serum of healthy young rats and the protective effect of H2O2on DNA damage induced by H2O2.
