摘　　要：目的通过检测泡球蚴感染小鼠M2型巨噬细胞特异性标志和基因表达水平的变化,观察M2型巨噬细胞的变化。方法将48只雌性BALB/c小鼠随机分为对照组及模型组。模型组利用保种BALB/c体内的囊泡制成注射用的悬液,通过腹腔注射完成泡球蚴感染模型的建立,对照组只注射生理盐水。于感染后90、180和300 d各取8只小鼠处死,剥离肝脏,一部分固定于甲醛溶液中,用于HE染色,采用免疫组化法检测组织CD163蛋白表达水平;另一部分冻存,采用qRT-PCR法检测Fizzl mRNA的相对表达量。结果与对照组相比,M2型巨噬细胞特异性标志CD163表达水平在感染90 d时升高明显,感染180 d时达到峰值,之后有下降趋势,但仍高(P均<0.05)。与对照组相比,M2型巨噬细胞特异性基因Fizz1 mRNA的相对表达量总体趋势和CD163的表达趋势相似(P均<0.05)。结论泡球蚴感染小鼠中、后期肝脏M2型巨噬细胞增多。Objective To observe the changes in M2macrophages by detecting the changes in M2-type macrophage-specific markers and gene expression levels in mice infected with alveolar echinococcosis.Methods Forty-eight female BALB/c mice were randomly divided into the control group and model group.In the model group,we used the vesicles in the BALB/c body to prepare a suspension for injection,and the infection model of the alveolar echinococcosis was completed by intraperitoneal injection.The control group was only injected with normal saline.Eight mice were sacrificed respectively at90,180and300d after infection,and the liver was removed.One part was fixed in formaldehyde solution for HE and immunohistochemical staining.The other part was frozen and the relative value of Fizzl mRNA was detected by qRT-PCR.Results Compared with the control group,the expression of M2macrophages'specific marker CD163increased significantly at90d,reached a peak at180d,and then decreased,but it was still high(P<0.05).Compared with the control group,the overall trend of M2macrophages'specific gene Fizz1mRNA expression was similar to that of its specific marker CD163(P<0.05).Conclusion The M2macrophages in the liver of the mice infected with alveolar echinococcosis increase in the middle and late stages.
