摘 要:目的探讨木犀草素对A375和B16F10黑素瘤细胞增殖、侵袭、凋亡及血管内皮生长因子(VEGF)表达的影响及其作用机制。方法将A375和B16F10细胞各分为4组:对照组、木犀草素20、40、60μmol/L组。CCK-8实验检测细胞增殖;Transwell实验检测细胞侵袭;流式细胞仪检测细胞凋亡;Western Blot检测Caspase-3、Bax、Bcl-2、VEGF、p-Akt、Akt、p-mTOR和m TOR的蛋白水平;RT-PCR检测VEGF的m RNA水平。结果与对照组相比,木犀草素呈浓度和时间依赖性抑制A375和B16F10细胞增殖;而且木犀草素组的细胞侵袭能力下降,细胞凋亡率升高,Bcl-2表达降低,而Bax和Caspase-3的表达增加。此外,木犀草素组的p-Akt和p-mTOR的蛋白水平显著下降。结论木犀草素能够抑制A375和B16F10细胞的增殖和侵袭能力,诱导细胞凋亡,这可能与降低VEGF表达,抑制PI3K-Akt-mTOR信号通路有关。Objective To determine the effects of luteolin on the proliferation,invasion,apoptosis and VEGF expression in A375 and B16F10 melanoma cells,and to investigate the underlying mechanisms.Methods A375 and B16F10 cells were divided into four groups respectively:control group,luteolin 20μmol/Lgroup,luteolin 40μmol/L group,luteolin 60μmol/L group.Cell proliferation was detected by CCK-8 assay.Cell invasion was detected by Transwell assay;Cell apoptosis was detected by flow cytometry.The protein levels of Caspase-3,Bax,Bcl-2,VEGF,p-Akt,Akt,p-mTOR and mTOR were detected by Western Blot.The mRNA level of VEGF was detected by RT-PCR.Results Compared with the control group,luteolin inhibited the proliferation of A375 and B16F10 cells in a concentration and time.dependent manner.The decreased invasive ability,the increased apoptosis rate,the decreased Bcl.2 expression,and the increased expression of Bax and Caspase-3 were found in the luteolin group.In addition,the protein levels of p-Akt and p-mTOR were significantly decreased in the luteolin group.Conclusion Luteolin can inhibit the proliferation and invasion,and induce apoptosis of A375 and B16F10 cells,which may be related to the decreased VEGF expression and the inhibition of PI3K-Akt-mTOR signaling pathway.
