摘  要:目的探讨自噬相关因子LC3及Beclin1在大鼠发育性髋关节发育不良早期退变关节软骨中的表达及意义。方法将80只新生大鼠随机分为模型组(40只)与对照组(40只),对模型组大鼠行双下肢并拢屈髋伸膝位固定以制造发育性髋关节发育不良模型。分别于2、4、6、8周龄时处死模型组和对照组大鼠,并测量这4个年龄段大鼠髋臼和股骨头的大小,通过苏木精-伊红(HE)染色及改良番红O-固绿染色观察髋关节软骨病理组织学改变,采用免疫组化及实时荧光定量聚合酶链式反应(PCR)检测软骨退变指标基质金属蛋白酶(MMP)-13、Ⅹ型胶原及自噬相关因子LC3和Beclin1在髋关节软骨中的表达与分布。结果模型组关节囊增厚,股骨头扁平,髋臼深度变浅,股骨头及髋臼长径与横径较对照组小。但随着年龄的增长,模型组股骨头及髋臼长径与横径仍呈增长趋势。6、8周龄时两组Mankins评分差异均有统计学意义(P<0.05)。2周龄时模型组关节软骨中Ⅹ型胶原表达量与对照组无明显差异,4、6周龄时模型组Ⅹ型胶原表达量均高于对照组(P<0.05);模型组关节软骨中MMP-13、LC3在各年龄段的表达量均高于对照组(P<0.05);2、4和6周龄时模型组Beclin1表达量高于对照组(P<0.05)。结论自噬在早期髋关节发育不良的髋臼软骨退变中起着重要作用。Objective To investigate the expression and significance of autophagy related factors LC3and Beclin1in the early cartilage degeneration of the developmental dysplasia of the hip.Methods80newborn SD rats were randomly divided into model group and control group,40newborn SD rats with straight-leg swaddle in model group.The rats were sacrificed at ages of2,4,6and8weeks,the size of acetabulum and femoral head were compared between model group and control group.Observing pathohistologic changes of articular cartilage by hematoxylin and eosinstain and improvement of safranin O-fast greenstaining.Immunohistochemistry and real-time fluorescence quantitative polymerase chain reaction(PCR)were used to detect the expression of collagenⅩ,matrix metalloprotein(MMP)-13,LC3and Beclin1in the early degeneration of articular cartilage.Results The capsule of necropsy specimens in the model group showed thicken and the femoral head was flat,the size of the femoral head and acetabulum were smaller than that in the control group.However,the length and diameter of femoral head and acetabulum in model group were increasing with age.There were significant differences in the Mankin score between groups at6,8weeks of age(P<0.05).There was no significantly difference in the expression of collagenⅩbetween two group at2weeks of age.At4,6weeks of age,the values of the collagenⅩof the model group were higher than that of the control group(P<0.05).At2,4,6,8weeks of age,the values of MMP-13,LC3in the articular cartilage of the model group were higher than that of the control group(P<0.05).The values of Beclin1in the articular cartilage of the model group were higher than that of the control group at2,4,6weeks of age(P<0.05).Conclusion Autophagy plays an important role in acetabular cartilage degeneration in early developmental dysplasia of the hip.
