摘  要:目的观察急性川崎病(KD)患儿外周血γδT细胞及IL-17A的变化,探讨产生IL-17A的γδT细胞在KD发病中改变及意义。方法分离人外周血单个核细胞(PBMCs),豆蔻酰佛波醇乙酯刺激后采用流式细胞仪检测γδT细胞和γδT17细胞占CD3+T细胞的百分比,同时采用ELISA测定血清IL-17A和TGF-β1水平,并比较两组间γδT、γδT17、IL-17A和TGF-β1的变化。结果相对于健康对照组,病例组患儿外周血γδT、γδT17、IL-17A和TGF-β1均明显增高[7.09±0.63 vs 11.24±2.31,1.25±0.53 vs 3.68±1.70,(4.41±1.09)ng/l vs (11.19±3.48)ng/L,(6.93±1.37)ng/ml vs (9.31±1.84)ng/ml](P<0.05)。结论γδT细胞在急性KD患儿高度活化,并且通过产生高水平细胞因子IL-17A介导了KD的炎症反应。Objective To observe the changes ofγδT cells and IL-17A in children with acute Kawasaki disease(KD),and to investigate the changes and significance of IL-17-producingγδT cells in the pathogenesis of KD.Methods The human peripheral blood mononuclear cells(PBMCs)were separated,and the percentage ofγδT cells andγδT17 cells in CD3+T cells was detected by flow cytometry after stimulation with PMA.At the same time,serum IL-17A and TGF-β1 levels were measured by using ELISA.The changes ofγδT,γδT17,IL-17A and TGF-β1 between the two groups were compared.Results Compared with the healthy control group,theγδT,γδT17,IL-17A and TGF-β1 in the peripheral blood of the case group were significantly increased[7.09±0.63 vs 11.24±2.31,1.25±0.53 vs 3.68±1.70,(4.41±1.09)ng/l vs(11.19±3.48)ng/L,(6.93±1.37)ng/ml vs(9.31±1.84)ng/ml](P<0.05).ConclusionsγδT cells are highly activated in children with acute KD,and may participate in the pathogenesis of KD by producing a high level of IL-17A.
